摘要
用高效液相色谱-二极管阵列检测器(HPLC-PDA)建立了苦荞中芦丁和槲皮素同时测定的方法;采用Intersil ODS-SP C18柱(150 mm×4.6 mm,5μm);流动相:甲醇-0.5%磷酸溶液(用磷酸二氢钾调节pH至4.5)(40:60),流速为1.0 mL/min,检测波长为255 nm。结果:芦丁和槲皮素峰均能良好分离,进样量的线性范围分别为1.98~39.60μg/mL和2.08~41.60μg/mL,平均加样回收率分别为99.2%和98.3%,RSD分别为0.3%和0.48%;试验结果表明所用方法简便、准确、重复性好,可用于苦荞中芦丁和槲皮素的含量测定。
Rutin and quercetin in Tartary buckwheat were analyzed using high performance liquid chromatography(HPLC)/photodiode array detection(PDA).An Intersil ODS-SP C18 column(150 mm×4.6 mm,5 μm) was used.The mobile phase consisted of methanol-0.5% phosphoric acid(adjusting pH to 4.5 by potassium dihydrogen phosphate)(40:60).The flow rate was 1.0 mL/min and the detection wavelength was at 255 nm.Result: The content of rutin and quercetin was liner over the range of 1.98~39.60 μg/mL and 2.08~41.60 μg/mL,respectively.The average recover was 99.2% and 98.3% with RSD of 0.3% and 0.48%,respectively.The results were: The method is simple,accurate,reproducible and can be used in the determining of rutin and quercetin tartary buckwheat.
