祛痹方对Ⅱ型胶原诱导的免疫性关节炎大鼠免疫调节因子的影响 被引量:3

Effect of Qubi Formula on the Immunomodulatory Factors in TypeⅡ Collagen-Induced Arthritis Rats

摘要 目的探讨祛痹方对Ⅱ型胶原诱导的免疫性关节炎(CIA)大鼠的作用及免疫调节机制。方法 SD大鼠72只随机分为空白对照组12只,模型组、雷公藤多苷组、祛痹方高剂量组、祛痹方低剂量组各15只。除空白对照组外其余各组采用尾根部皮下注射Ⅱ型胶原与不完全弗氏佐剂的方法诱导出CIA模型,造模第15天祛痹方高、低剂量组分别给予祛痹方混悬液6.66g/(kg·d)、3.33g/(kg·d),雷公藤多苷组给予雷公藤多苷混悬液9.68mg/(kg·d),空白对照组和模型组大鼠等体积纯净水,各组每天灌胃1次,连续4周。观察各组大鼠关节病理损伤并进行半定量评分,检测血浆中抗Ⅱ型胶原抗体(COL-2)、白细胞介素-1β(IL-1β)、白细胞介素6(IL-6)含量。结果模型组大鼠膝关节病理积分,血浆COL-2、IL-1β、IL-6含量较空白对照组明显升高(P<0.05或P<0.01);与模型组比较,祛痹方高剂量组与雷公藤多苷组大鼠关节病理积分,血浆COL-2、IL-1β、IL-6含量明显降低(P<0.05或P<0.01);祛痹方高剂量组大鼠关节病理积分,血浆COL-2、IL-1β水平亦明显低于雷公藤多苷组和祛痹方低剂量组(P<0.05或P<0.01)。结论高剂量祛痹方显著减轻了CIA大鼠膝关节损伤,其作用机理可能与降低血浆COL-2水平,下调血浆IL-1β、IL-6含量有关。 Objective To study the therapeutic effect and Immunomodulatory mechanism of Qubi Formula (Impediment Dispelling Formula) for type Ⅱ collagen induced arthritis (CIA) in rats. Methods Totally 72 SD rats were randomized into the Blank control group, model group, tripterygium glycosides group, Quhi Formula high-dose group and Quhi Formula low- dose group, with 12, 15, 15, 15 and 15 in each. The CIA models were induced by subcutaneous injection of collagen type Ⅱ and incomplete Freund's adjuvant on the tail root of rats in all groups except the blank control group. On the 15th day of modeling, the Qubi Formula high-dose and low dose groups were gavaged with 6.66g/(kg · d) and 3. 33g/(kg · d) Qubi Formula suspension, the tripterygium glycosides group was gavaged with 9.68 mg/(kg · d) tripterygium glycosides suspension and the blank control group and model group were garaged with equal volume of pure water once daily for 4 weeks. The pathological damage of joints in each group was observed and semi-quantitative score was calculated. The contents of anti type Ⅱ collagen antibodies (COL-2), interleukin-1β (IL-1β) and interleukin 6 (IL-6) were detected. Results Comparing with the blank control group, the score of knee joint pathology and contents of COL -2, IL-1β and IL -6 were significantly increased in the model group (P〈0.05 or P〈0. 01). Comparing with the model group, the score of knee joint pathology and contents of COL- 2, IL-1β and IL-6 were significantly decreased in the Qubi Formula high-dose group and tripterygium glycosides group (P〈 0.05 or P〈0.01). Comparing with the Qubi Formula low-dose group and tripterygium glycosides group, the score of knee joint pathology and contents of COL-2 and IL-1β were significantly decreased in the Qubi Formula high-dose group (P〈0.05 or P〈0.01). Conclusion High-dose Qubi Formula can obviously alleviate knee injury in CIA rats that may be relevant to reducing the COL-2 level and down regulating the contents of IL-1β and IL-6.
