促育生精颗粒对环磷酰胺致少弱精症模型大鼠的影响 被引量:10

Effect of Cuyu Shengjing granules on cyclophosphamide(CP)-induced asthenospermia and oligozoospermia in model rats

摘要 目的:探讨促育生精颗粒(Cuyu)对环磷酰胺诱导的少弱精症模型大鼠精子活力的改善作用及相关机制。方法:50只雄性Wistar大鼠,随机分成正常对照组,模型组,低、中、高剂量中药治疗组。模型组,低、中、高剂量治疗组腹腔注射环磷酰胺(CP)30 mg·mL-1·d-1,共5 d。建立模型后,高、中、低剂量治疗组大鼠分别按生药量3.37×104,1.69×104,0.84×104 mg·kg-1·d-1剂量给药,每只大鼠灌胃3 mL促育生精颗粒药液,持续35 d。普通光学显微镜观察附睾精子数目、精子活力;流式细胞术检测精子凋亡率;HE染色观察生精小管层数。结果:各剂量治疗组与模型组比较精子数目增加(P<0.05)、精子凋亡率下降(P<0.05)、生精小管层数增加(P<0.05),中剂量、高剂量治疗组与模型组比较a级精子比率上升(P<0.05)。结论:通过促育生精颗粒治疗可以增加少弱精模型大鼠精子数,提高精子活力,此作用可能与降低精子凋亡率,增加生精小管生殖细胞层数有关。 OBJECTIVE To study the effect of Cuyu Shengjing granule(Cuyu) in improving sperm activity on model rats of the cyclophosphamide (CP)-induced asthenospermia and oligozoospermia and its possible mechanisms. METHODS 50 male Wistar rats were randomly divided into control group, model group, and low-dose, middle-dose and high-dose treatment group. The rats of the model and treatment groups were given with CP of 30 mg.kg^-1 .d^-1 through intraperitoneal injection for 5 days. The low-dose , middle-dose and high dose groups were given with 3 mI. medicinal liguid of Cuyu Shengjing granule which equal to 0.84× 10^4 mg·kg ^-1·d 1 , 1.69× 10^4 mg·kg^-1 ·d^- 1 ,3.37 ×104 mg·kg^- 1 ·d ^-1 by gastric gavage. After 35 days, sperm mobili- ty rate, sperm amount were measured by light microscope. Sperm apoptosis rates were detected by flow cytometry, and the numbers of seminiferous tubule layer were observed by HE staining. RESULTS Compared with model group, the numbers of sperm of every treatment groups were increased (P〈0. 05), apoptosis rates of the sperm were significantly decreased (P〈 0. 05), the numbers of seminiferous tubule layer were increased(P〈0. 05), and the sum of A and B degree sperm rate were in creased, but the low dose group had no difference. CONCLUSION The number of sperm and the activity of sperm of model rats of the cyclophosphanfide(CP) induced asthenospermia and oligozoospermia were increased by treatment of Cuyu Shengjing granul. The effects may be related to restraining sperm from apoptosis, and increasing the number of seminiferous tubule layer.
