摘要
目的 探讨内皮细胞分化基因-1(EDG-1)、丝切蛋白-1(CFL-1)在原发性肝细胞癌(HCC)中的表达水平及其与临床病理特征和两者之间的相互关系.方法 采用免疫组织化学方法,检测EDG-1、CFL-1在57例原发性肝细胞癌组织(实验组)及15例癌旁组织(对照组)中的表达,并与临床病理资料进行比较分析.随访所有病例术后的生存时间,并进行生存分析.结果 EDG-1 、CFL-1阳性表达分别为54.4% (31/57)和66.7% (37/57),均明显高于正常肝组织的表达,差异有统计学意义(P<0.05),且呈正相关(P<0.05).EDG-1、CFL-1在肿瘤转移组及低分化组中的表达阳性率明显高于无肿瘤转移组及高+中分化组,差异有统计学意义(P<0.05).COX多因素回归分析的结果提示,肿瘤转移、癌栓、病理分型、EDG-1及CFL-1的表达为影响预后的独立因素.结论 EDG-1 、CFL-1可能作为判断HCC恶性程度的预测指标,其表达水平有助于评价肝细胞癌患者的预后.
Objective To explore the correlation between the expression of endothelial differentiation gene 1 (EDG-1) and cofilin-1 (CFL-1) in hepatocellular carcinoma (HCC) with clinicopathological characteristics.Methods Immunohistochemistry was applied to detect the differential expression of EDG-1 and CFL-1 in 57 tumor specimens (experimental group) and 15 paracancerous hepatic tissue specimens (control group).The survival time of all cases after surgery was followed up.Results The immunohistochemistry showed that immunostaining of EDG-1 and CFL-1 was located in the cytoplasm,and the positive rate was 54.4% (31/57) and 66.7% (38/57) respectively in experimental group,significantly higher than 13.3% (2/15) in control group (both P < 0.05).There was a positive correlation between the expression score of EDG-1 and CFL-1 in HCC tissues (P < 0.05).The clinicopathological evaluation revealed that the expression of EDG-1 and CFL-1 was significantly higher in HCC cases with metastasis than in those without metastasis (P < 0.05),and correlated with differentiation degree of HCC tissues (P <0.05).Cox regression analysis showed that metastasis,cancer embolus,differentiation degree and the expression of EDG-1 and CFL-1 were independent prognostic factors (P < 0.05).Conclusion EDG-1 andCFL-1 could be an indication to identify the malignant degree of HCC.EDG-1 and CFL-1 are likely to be valuable indications in evaluating of the prognosis of patients with HCC.
