调节性T细胞在乳腺癌患者体内的表达及其临床意义 被引量:5

Expression and significance of regulatory T cells in patients with breast cancer

摘要 目的 探讨乳腺癌患者外周血和肿瘤组织CD4+ CD25+ CD127low/-T细胞(Tregs)的变化及其临床意义.方法 采用流式细胞术对48例乳腺癌患者及12例健康志愿者外周血Treg细胞进行检测,同时对10例乳腺癌患者癌组织及癌旁组织Treg细胞进行检测.结果 10例乳腺癌患者癌组织Treg细胞表达为(39.74 ±9.02)%,显著高于其癌旁组织的表达[(29.81±8.52)%,P<0.05].48例乳腺癌患者外周血Treg细胞表达为(7.04±1.56)%,显著高于12例健康志愿者外周血Treg细胞的表达[(4.01±1.05)%,P<0.05].且随着疾病的进展,外周血Treg细胞水平升高,Ⅱ期+Ⅲ期乳腺癌患者外周血Treg细胞表达为(8.15±1.28)%,显著高于Ⅰ期患者[(6.39±0.81)%,P<0.05]和健康志愿者[(4.01±1.05)%,P<0.05].结论 Treg细胞在乳腺癌患者外周血和肿瘤组织的高表达,可能是肿瘤长期免疫逃逸的重要原因,其外周血数量与乳腺癌进展密切相关,越晚期表达水平越高. Objective To investigate the expression and clinical signifycance of CD4 + CD25 + CD127low/-regulatory T cells (Tregs) in peripheral blood and tumor tissue of breast cancer patients.Methods The percentage of Tregs in tumor tissues,and adjacent tissues from 10 patients with breast cancer,peripheral blood from 48 patients with breast cancer and 12 healthy volunteers was detected by using flow cytometry.Results The percentage of Tregs in 10 breast cancer patients was (39.74 ± 9.02) % in tumor tissues,significantly higher than that in the adjacent tissues [(29.81 ± 8.52)%,P <0.05].The percentage of Tregs in peripheral blood of 48 breast cancer patients was (7.04 ± 1.56) %,significantly higher than in 12 healthy volunteers [(4.01 ± 1.05) %,P < 0.05].With the progress of the disease,the proportion of Tregs in peripheral blood of breast cancer patients had a higher level.The levels of Tregs in peripheral blood of breast cancer patients with stage (Ⅱ + Ⅲ) were signifycantly higher than those of patients with stage Ⅰ [(6.39 ± 0.81) %,P < 0.05] and heathy volunteers [(4.01 ± 1.05) %,P < 0.05].Conclusion There is high expression of Tregs in peripheral blood and tumor tissues of patients with breast cancer,which might be closely related to long-term immunoescape of tumor.Besides,the populations of Tregs in peripheral blood may contributed to the progression of breast cancer.
