摘要
目的研究人脱细胞膀胱黏膜下基质(AUBS)异种移植的转归及免疫原性。方法将人AUBS异种移植于SD大鼠的腹壁肌肉内,分别于术后3、6、10、14、21和28 d取材进行组织学观察和FITC标记的山羊抗大鼠IgG免疫荧光染色观察。将人脱细胞真皮基质和未脱细胞膀胱黏膜下层作为对照进行对比观察。结果人AUBS可以诱导组织细胞浸润、生长,14 d内基质移植区血管化,28 d内移植基质被机体降解吸收。IgG免疫荧光染色观察显示,基质移植后局部无明显IgG免疫复合物的沉积。结论人AUBS植入大鼠腹壁肌肉后可以诱导组织细胞长入和血管化,并在新生组织形成过程中迅速降解吸收,不引起明显的免疫反应。
Objective To investigate the immunogenicity and consequences of heterogenic transplantation of acellular urinary bladder submucosa(AUBS).Methods Human AUBS was transplanted into abdominal muscle of SD rats.The untreated human AUBS served as control.Samples were taken at 3 d,6 d,10 d,14 d,21 d and 28 d after transplantation.Histological and immunological evaluation was performed on the samples.Results Human AUBS could induce cell infiltration and growth after transplantation.Vessel infiltration was revealed at 14 d.The transplanted matrix was degraded and absorbed at 28 d.FITC labelled goat anti-rat IgG immunofluorescence revealed no obvious deposition of IgG immune compound.Conclusion Human AUBS can induce cell and vessel infiltration in the abdominal muscle of SD rats.It degrades as new tissues are formed,but does not induce obvious immunological reactions.
