鼻敏片对变态反应性鼻炎豚鼠黏附分子的影响

Effects of Bimin Tablet on Adhesion Molecules Expression in Guinea Pigs with Allergic Rhinitis

摘要 目的:观察鼻敏片干预变态反应性鼻炎豚鼠黏附分子的表达及黏附分子之间的相关性,以探讨其作用机制。方法:将豚鼠按卵清蛋白致敏法制备变态反应性鼻炎模型,随机分为空白组、模型组、鼻敏片低剂量组、鼻敏片高剂量组、开瑞坦组,每组12只,给药15天。检测豚鼠血清中细胞黏附分子-1(ICAM-1)及其配体淋巴细胞相关抗原-1(LFA-1),以及血管细胞间黏附分子-1(VCAM-1)及其配体很晚抗原-4(VLA-4)。结果:血清ICAM-1、LFA-1、VCAM-1、VLA-4在各组中的表达比空白组高,差异均有显著性意义(P<0.05),血清ICAM-1、LFA-1、VCAM-1、VLA-4在各组中的表达比模型组低,差异均有显著性意义(P<0.05),血清ICAM-1、LFA-1、VCAM-1、VLA-4在鼻敏片低剂量组、鼻敏片高剂量组的表达与开瑞坦组比较,差异无显著性意义(P>0.05)。血清中ICAM-1与LFA-1的含量经分析后,证实呈线性回归关系(r=0.812,F=93.078,P=0.000)。血清中VCAM-1与VLA-4的含量经分析后证实呈线性回归关系(r=0.733,F=71.491,P=0.000)。结论:鼻敏片能降低变应性鼻炎豚鼠血清中ICAM-1、LFA-1、VCAM-1、VLA-4的含量。 Objective. The effects of Bimin tablet(BMT)on expression of adhesion molecules in guinea pigs with allergic rhinitis and the correlation between the adhesion molecules were investigated for the exploration of the therapeutic mecha- nism of BMT. Methods: The guinea pigs models were established by ovalbumin(OVA), and then were randomly divide into blank group, model group, Loratadine group and low-, high-dose BMT groups. Each group had 12 guinea pigs, and the medication lasted 15 days. Before and after the medication, we observe the contents of intercellular adhesion molecule(I- CAM-l) and its Iigand lymphocyte function-associated antigen-1 (LFA-1), vascular cell adhesion molecule-1 (VCAM-1) and ks ligand very late antigen-4(VLA-4) in the serum of guinea pigs by enzyme-linked immunospecific assay(ELISA). Results: The contents of ICAM-1, LFA, VCAM-1 and VLA-4 were increased obviously in the model group and medication groups compared with the blank group(P 〈 0.05) before treatment, and were decreased in the medication groups after treatment(P 〈 0.05 compared with those in the model group). The differences of ICAM-1, LFA, VCAM-1 and VLA-4 contents were in- significant between BMT groups and Loratadine group(P 〉 0.05). Linear regression relationship was shown between ICAM-1 and LFA(F=93.078, P=0.000)and between VCAM-1 and VLA-4(~=71.491, P=0.000). Conclusion: Bimin tablet can decrease the serum contents of ICAM-1, LFA, VCAM-1 and VLA-4 in guinea pigs with allergic rhinitis.
