摘要
目的观察脂多糖所致的血管外膜损伤对大鼠颈动脉收缩功能及心肌素等因子表达的影响。方法利用脂多糖(LPS)刺激大鼠颈动脉外膜;采用不同浓度的5羟色胺诱发血管痉挛;采用HE染色法观察颈动脉病理改变;Western blot检测颈动脉中心肌素的变化;实时定量PCR法检测颈动脉中miR-145及miR 1的表达。结果 LPS处理后大鼠颈动脉血管中膜明显增厚,平滑肌细胞数量增多、排列紊乱。应用0.2%的5羟色胺时,模型组动脉血流量下降程度明显大于对照组。模型组心肌素蛋白的表达水平明显高于对照组(P<0.05),miR-145和miR-1的表达水平明显低于对照组(P<0.05)。结论本研究应用LPS刺激大鼠颈动脉血管外膜第5天动脉中膜平滑肌增生,并对5羟色胺的收缩反应增强。模型组血管平滑肌中心肌素的蛋白表达上调,miR 145和miR 1的表达下调。
Objective To investigate the therapeutice efficacy of anterior vaginal wall flap for the treatment of SUI.Methods A total of 44SUI patients admitted in our department were enrolled in this study,and randomized to two groups.SUI operation using wall anterior vaginal flap as suspending material and upgraded based on Urethral suspension surgery was performed in 22 patients (experiment group),and Urethral suspension surgery was carried out in another 22 patients (control group).The general condition,quality of life and ttrodynamic study after (follow-up of 2 months and one year) operation were compared between two groups.The cure rate was statistically analyzed.Results After 2 months of follow-up survey,among the experimental group,19 patients with urinary incontinence symptoms were completely cured,2 cases still have symptoms of mild urinary incontinence,1 case without obvious symptoms improvement.The cure rate was 86.36%,22 patients had no postoperative surgical site infection,bladder injury,hematoma,postoperative complications such as vaginal foreign body exposure.In the control group,21 patients were completely cured with a rate of 95.45%,there was only 1 patient still had dysuria symptoms ; during the 1 year' s follow-up survey,the cure rate was 95.45% of the experiment group.None of them had complications such as infection,bladder injury,hematoma,foreign body exposure.In the control group,20 cases in 22 were cured completely with a cure rate of 90.91%.1 case accidentally had misnicturition,and 1 case received mesh remove caused by the mesh invasion.Conclusion Homograft made by PAVF showed a clear effect,which was safe with less complication compared with Urethral suspension surgery as a new technique.
