摘要
目的探讨CYP2J2基因多态性和吸烟的交互作用及其与心肌梗死(MI)的相关性。方法采用聚合酶链反应-限制性片段长度多态性方法,对85例MI患者(MI组)和85例健康体检者(对照组)CYP2J2基因标签SNP进行分析,并分析其多态性和吸烟的交互作用及其与MI的关系。结果 CYP2J2基因型分布符合Hardy-Weinberg平衡;TT、GT、GG基因型频率在MI组中分别为3.5%、4.7%、91.8%,在对照组为1.2%、3.5%、95.3%,两组之间基因型分布差异无统计学意义(P>0.05);CYP2J2基因多态性和吸烟的交互作用分析显示,以GG基因型(野生型)不吸烟者作为基线(OR=1),则GG基因型吸烟者发生MI的风险增加1倍(OR=2.0),而携带非GG基因型(突变型)的吸烟者发生MI的风险增加了7.7倍(OR=8.7);Logistic回归分析结果显示,在调整了年龄、性别、高血压、糖尿病、高胆固醇血症等传统危险因素之后,吸烟和CYP2J2基因多态性之间的交互作用是MI发生的重要危险因素。结论 CYP2J2基因多态性和吸烟的交互作用增加MI的风险。
Objective To investigate the correlation between the cooperating effect of Cytochrome P2J2(CYP2J2) gene polymorphism and smoking and its impact on myocardial infarction( MI).Methods A total of 85patients with MI and 85 healthy subjects were enrolled in this study.The CYP2J2 gene tagging SNP was analyzed by polymerase chain reaction-restriction fragment length( PCR-RFLP).The interaction of CYP2J2 gene polymorphism and smoking and its correlation with MI was analyzed.Results The genotype distributions in control group and MI group were all in the Hardy-Weinberg equilibrium.The frequencies of TT,GT,GG genotype were 3.5%,4.7% and91.8%,respectively in MI group; the frequencies of TT,GT,GG genotype were 1.2%,3.5% and 95.3%,respectively in control group; there was no significant difference in frequencies of three genotypes between two groups(P0.05).The analysis of the interaction between CYP2J2 gene polymorphism and smoking showed that taking the GG genotype( wild type) as baseline(OR=1),the risk of MI increased by two fold(OR=2.0) in smoker of GG genotype,while the risk of MI increased by 8.7 fold(OR=8.7) in smoker carrying non-GG genotype( mutant).Logistic regression analysis showed that after adjusting traditional risks such as age,sex,hypertension,diabetes mellitus and hypercholesterolemia,the interaction of smoking and CYP2J2 gene polymorphism is the main risk factor of MI.Conclusions The interaction between CYP2J2 gene polymorphism and smoking may increase the risk of MI.
