微量全血培养在HIV感染诊断中的应用

Diagnosis of Human Immunodeficiency Virus Infection by Small Volumes of Whole Blood Culture

[目的 ]建立一种简便、有效的 HIV感染检测方法 ,用于儿童 HIV垂直感染的早期诊断及可疑感染者的确认。 [方法 ]用肝素化全血 (每孔分别 10 0 0、 5 0 0、 2 5 0、 12 5、 6 2 .5、 10 μl)与正常人激活的 PBMC10 6在 2 4孔板中进行复孔共培养或与新鲜的正常人 PBMC同时刺激共培养来分离 HIV病毒 ,培养上清液用 P2 4抗原 EL ISA试剂盒做检测。 [结果 ]4例全血培养中分离出 3株 HIV- 1,微量全血培养出现阳性的时间顺序为 10 μl>6 2 .5 μl>12 5 μl>2 5 0 μl>5 0 0 μl>10 0 0 μl。 [结论 ]微量全血培养有较高的敏感性和特异性 ,能够用少量全血做出快速诊断 ,可应用于 HIV- 1垂直感染的早期诊断。

Objective] To develop an easier and efficient method for the early diagnosis of HIV vertical infection and the identification of HIV suspected infection case [Methods] Heparinized whole blood collected from HIV/AIDS cases was co cultured with duplication in 24 well plates (1000、500、250、125、62 5、10 μl/well) with PHA P stimulated 10 6 PBMC from normal donors or with fresh PBMC that stimulated in the same time to isolate HIV Cultured supernatant was detected by Vironostika HIV 1 Antigen Microelisa System [Results] Three strains of HIV 1 were isolated from 4 cultures of whole blood The time sequence of showing positive result was 10 μl>62 5 μl>125 μl>250 μl>500 μl>1000 μl [Conclusion] Small volumes of whole blood culture is of higher sensitivity and specificity and can be used for the early diagnosis of HIV vertical infection
