摘要
目的研究氯化锦葵色素-3-O-葡萄糖苷(Mv-3-glc-Cl)对细胞肿瘤坏死因子(TNF-α)诱导人脐静脉内皮细胞(HUVEC)所产生炎症的抑制作用。方法实验分为,空白组(DMSO)、10 g/L T NF-α刺激组、浓度为1、10、50、100 mol/L MV-3-gle-Cl+10 g/L TNF-α刺激组,通过ELISA方法检测细胞上清中可溶性单核细胞趋化蛋白(MCP-1)蛋白含量的变化;用实时荧光定量PCR检测细胞中MCP-1 mRNA表达量变化;Western blot检测IκBα蛋白含量的变化;通过免疫荧光方法测定核转录因子κB的核异位变化。结果 Mv-3-glc-Cl呈浓度依赖性的抑制细胞肿瘤坏死因子(TNF-α)诱导人脐静脉内皮细胞(HUVEC)MCP-1蛋白表达,其抑制作用可能与NF-κB信号通路有关。结论 Mv-3-glc-Cl对TNF-α诱导HUVEC的炎症反应起到抑制作用。
Objective To study the effect of malvidin-3-glucoside-chloride (Mv-3-glc-C1) on TNF a-induced inflammation in human umbilical vein endothelial cell (HUVEC). Methods HUVEC were treated with TNF a (10 gg/L), TNF ct and Mv-3-glc (1, 10, 50, 100 gmol/L), respectively. DMSO was used as control. The soluble monocyte chemoattractant protein-1 (MCP-1) content in cellular supernatant was detected by EL1SA. The changes of mRNA expression level of MCP-1 in cells were detected by RT-PCR. The protein of~ B was assessed by Western-blot. The activity of NF-~B was evaluated by immunofluorescence. Results Mv-3-glc-C1 appeared to specifically down-regulate the TNF u-induced MCP-I protein expression in HUVEC in a concen- tration-dependent manner. Inhibition of Mv-3-glc-C1 on protein expression might be related to NF-nB suppression in HUVEC. Conclusion Mv-3-glc-C1 can inhibit TNF a-stimulated inflammation in HUVEC.
