摘要
采用循环伏安法、光谱法和粘度实验研究了硫脲壳聚糖铜配合物与鲱鱼精DNA的相互作用。结果发现:随硫脲壳聚糖铜浓度的增加,DNA的紫外吸收光谱呈增色效应和红移;硫脲壳聚糖铜离子在电极上的反应主要由扩散过程控制,加入DNA后氧化还原峰电流降低,且式量电位正移,说明硫脲壳聚糖铜与DNA以嵌入方式生成一种非电活性超分子复合物,导致硫脲壳聚糖铜配合物的峰电流降低。实验结果表明硫脲壳聚糖铜与DNA的相互作用模式为嵌插作用。
In this study,the interaction of thiourea-chitosan-Cu and herring sperm DNA was investigated using cyclic voltammetry,UV-Vis method and viscostiy measurement.It was found that the ultraviolet absorption intensity of DNA increased as the concentration of thiourea-chitosan-Cu increased.The electrochemical results showed that the electrochemical reactions of the thiourea-chitosan-Cu on the glass carbon electrode were controlled by the process of diffusion.Adding DNA into the thiourea-chitosan-Cu solution resulted in the positive shift of the cathodic peak potential and the decrease of cathodic peak,illuminated there was a super-molecular complex produced.The result suggested that the thiourea-chitosan-Cu bind to DNA by intercalative action and static electricity.
