摘要
目的:建立大鼠血浆中冬凌草乙素的高效液相色谱(HPLC)分析方法,测定冬凌草乙素的血浆蛋白结合率,为临床安全用药提供参考。方法:采用超滤法结合HPLC法测定冬凌草乙素与大鼠血浆蛋白的结合率。结果:当冬凌草乙素在大鼠血浆中浓度为100,600,3 200 ng·mL-1时,冬凌草乙素与大鼠血浆的蛋白结合率分别为(70.9±3.4)%,(71.0±3.0)%,(69.6±2.9)%。结论:冬凌草乙素与大鼠血浆蛋白具有中等强度的结合,并且在考察浓度范围内血浆蛋白结合率无浓度依赖性。
Objective:To establish an HPLC analysis method for determination of ponicidin in rat plasma and to determine the plasma protein binding rates of ponicidin for providing information on the safe use of medication. Methods:The protein binding rates of ponicidin in rat plasma samples were determined by ultrafiltration and HPLC. Results:The plasma protein binding rate of ponicidin with rat plasma were (70.9 ± 3.4)%, (71.0 ± 3.0) % ,and (69.6 ± 2.9)% at the concentration of 100,600, and 3 200 ng. mL^-1, respectively. Conclusion: Ponicidin has medium binding ability to plasma protein. The method has good repeatability,sensitivity, specificity, and simpleness, thus fulfills the requirement. The plasma protein binding of ponicidin was independent of the investigated concentrations.
