摘要
本文提出用差分脉冲极谱法直接测定赖氨酸或精氨酸。它是基于在硼酸钠缓冲溶液中赖氨酸或精氨酸与乙醛发生化学反应,其反应产物的峰电位比其它氨基酸的峰电位较正100~150mV。对赖氨酸而言,蜂电位为-1.50V;浓度线性范围为8×10^(-4)~1.0×10^(-6)mol/L;检测限为8.0×10^(-7)mol/L。而对精氨酸而言,则分别为-1.50V;1.0×10^(-3)~2.8×10^(-6)mol/L;1.4×10^(-6)mol/L。在试样测定中,获得满意的结果。
A differential polarographic method for the determination of lysine and arginine is cescribed. It is based on the reaction between lysino or arginine and acetaldehyde in a borax buffer solution (pH=10.10). The peak potential of the reaction product is-1.50V(vs.SCE) which is about 100~150mV more posit ve than that of other amino acids. The peak height is proportional to the concentration of lysine over the range of 1×10^(-6)~8×10^(-4) mol/L with a detection limit of 8×10^(-7) mol/L. Whereas for arginine, they are 2.8×10^(-4)~1×10^(-3)mol/L and 1.4×10^(-6) mol/L respectively. The method has been applied to the determination of them in some food and serum samples with good results.
