远位触液神经元Fos蛋白的表达在CCI致神经病理性疼痛中的作用 被引量:1

摘要 目的 :研究坐骨神经松扎致神经病理性疼痛大鼠远位触液神经元Fos蛋白表达的变化。方法 :雄性SD大鼠16只,随机分为2组(n=8):假手术组、坐骨神经松扎模型(chronic contriction injury,CCI)组。采用侧脑室注射霍乱毒素B亚单位结合辣根过氧化物酶(cholera toxin subunit B labeled with horseradish peroxidase,CB-HRP)和CB-HRP/Fos免疫组织化学双重标记技术,观察大鼠远位触液神经元的Fos表达的变化。结果:各组大鼠都可见大量的CB-HRP标记神经元,组间比较差异无统计学意义(P>0.05)。CCI术前该细胞簇内未见有CB-HRP/Fos双重标记神经元,CCI术后可见CBHRP/Fos双重标记神经元,尤其术后5、7、14 d,CB-HRP/Fos双重标记神经元数目显著增多(P<0.01)。与假手术组比较,CCI组1、3 d CB-HRP/Fos双重标记的神经元数目增加(P<0.05),CCI术后5、7、14 d CB-HRP/Fos双标的神经元明显增加(P<0.01)。结论:远位触液神经元可能参与坐骨神经松扎致神经病理性疼痛的形成和维持。 Objective: To study the expression changes of Fos protein in chronic constriction injury(CCI)-induced neuropathic pain at distal cerebrospinal fluid contacting neurons in rats. Methods: Male SD rats were randomly divided into one of two groups(n=8): sham operation group, CCI group in a rat model of neuropathic pain induced by CCI. Using icy CB-HRP and CB-HRP/Fos immunohistochemical double labeling technique, we observed the expression of Fos protein in rat distal fluid contacting neurons. Results: The rats were seen in a large number of CB-HRP labeled neurons,the difference between groups is not statistically significant(P〉0.05). When no CCI stimulation, there were no CB-HRP/Fos labeled neurons in rat definite region, compared with preoperative CCI. After CCI 1, 3 d, there were increasing of double-labeled of CB-HRP/Fos CSF-CNs in CCI group as compared to that of sham group(P〈O.05). After CCI 5, 7, 14 d,there were conspicuous increasing of double- labeled of CB-HRP/Fos in CCI group as compared to sham group (P〈0.O1). Conclusion: The distal cerebrospinal fluid contacting neurons involved in the generation and maintence of the neuropathic pain induced by chronic constriction injury of the sciatic nerve.
