摘要
目的探讨RASSFIA基因对胃癌细胞自噬的影响情况及其可能的机制。方法通过基因重组技术已构建了高表达RASSF1A基因的人胃癌SGC-7901细胞株,然后采用聚合酶链反应(PCR)技术检测SGC-7901细胞转染RASSF1A基因前后自噬BECLIN-1基因mRNA表达情况,并使用流式细胞仪检测SGC-7901细胞转染RASSF1A基因前后凋亡情况。结果胃癌SGC-7901细胞转染了RASSF1A基因后,凋亡增加,BELINE 1表达下降。结论 RASSFIA基因对胃癌细胞自噬有负调控作用。
Objective To explore the effect of RASSFIA gene on the autophagy of gastric cancer cells and its mechanism. Methods A high expression of RASSF1A gene in human gastric cancer cell line of SGC-7901 was built through gene recombi nant technology, polymerase chain reaction (PRC) technology was adopted to detect the expression of mRNA in autography BE- CLIN-1 gene before and after transfection with RASSF1A gene in SGC-7901, and flow cytometry (FCM) was applied to detect the apoptosis of SGC-7901 cells before and after transfection with RASSFIA gene. Results After trasnfecting with RASSF1A gene, the apoptosis of SGC-7901 cells in gastric cancer increased ,and the expression of BELINE1 decreased. Conclusion RASSFIA gene has a negative regulatory role on the autophagy of gastric cancer cells.
