摘要
目的基于UPLC-代谢组学技术研究茯苓皮结合有氧运动对高脂血症大鼠的影响。方法雄性SD大鼠,分为5组,每组8只。高脂饲料喂饲大鼠4周,同时进行4周的茯苓皮、有氧运动、茯苓皮结合有氧运动治疗。实验第1天和第2周末尾静脉取血,第4周后处死动物,采用UPLC-代谢组学方法推测高脂血症模型是否成功。比较血清TG、TC、HDL-C及LDL-C含量,同时测定血清CAT、GSH-Px、MDA和SOD的浓度。结果在4周时间,代谢组学和生化指标证明高脂血模型成功建立,不同时间段产生不同的损伤程度且明确了高脂血症模型的时间进展轨迹。生化指标显示茯苓皮和有氧运动均可显著降低大鼠血清TG、TC和LDL-C提高、HDL-C水平;茯苓皮结合有氧运动能显著升高大鼠血清GSH-Px、SOD、CAT水平,同时降低MDA的水平,脂质过氧化产物水平显著降低。结论(1)代谢组学结果显示高脂血症模型已成功建立;(2)茯苓皮结合有氧运动可治疗高脂血症大鼠血脂异常,并有较明显的抗脂质过氧化作用;(3)代谢组学可以作为有氧运动治疗疾病程度及效果的一个新的判断方法。
Objective Metabolomics study on metabolic disorders in a hyperlipemia rat model and intervention effects of Poria cocos peel in combination with aerobic exercise in rats using UPLC method. Methods Male SD rats were divided into five groups (n = 8 rats/group). The rats were raised by high fat diet for four weeks; meanwhile these rats were treated with Poria cocos peel and aerobic exercise for four weeks. The blood samples were collected from retro - orbital puncture with ether and transferred into tubes on the first day and the second weekend. Then those rats were executed, and the model of hypenlipemia was then established and the model was verified by UPLC. The serum TG, TC, HDL - C, LDL - CTC, and GSH - Px were detected, at the same time the SOD, CAT and MDA concentrates also be measured. Results According to metabolomic method and biochemical parameters, the hyperlipemia rat model was successfully established in four weeks. The results showed the time progress of metabolic disorders in the different periods. The results showed that Poria cocos peel and aerobic exercise could decrease the concentration of serum LDL - C, TC and TG remarkably and raise the concentration of HDL - C. The activity of SOD, GSH - Px and CAT in Poria cocos in combination with aerobic exercise group is promoted and the level of MAD is decreased distinctly. Conclusions ( 1 ) The metabolomic result showed the model of hypenlipemia was then estab-lished. (2) Poria cocos peel in combination with aerobic exercise could treat the abnormity of serum lipid and has obviously anti - lipoperoxidation effect. (3) Metabolomics could also be a new method for studying on the therapeutic effect of aerobic exercise.
