摘要
目的:研究槲皮素对高糖诱导的小胶质细胞炎性反应的改善作用。方法:将体外培养的原代大鼠小胶质细胞分成对照组、高糖组、槲皮素低、中、高(3,10,30μmol·L-1)剂量组。采用ELISA测定培养液上清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α),白介素-1β(interleukin-1β,IL-1β);采用RT-PCR测定TNF-α、IL-1β、诱导型一氧化氮合成酶(inducible nitric oxide syntheses,iNOS)mRNA表达;采用western blot检测Toll样受体4(Toll-like receptor 4,TLR4)蛋白表达。结果:槲皮素显著地抑制了高糖诱导小胶质细胞中TNF-α、IL-1β、iNOS mRNA的表达,并显著得降低了TNF-α、IL-1β的释放,同时降低了TLR4的表达。结论:槲皮素可通过抑制高糖对小胶质细胞的激活,具有较好的中枢抗炎作用。
OBJECTIVE To study the inhibitory effect of quercetin on high-glucose induced inflammatory reaction in primary microglia cells of rats. METHODS High-glucose induced inflammatory reactions in primary microglia cells of rats were treated with quercetin (3, 10, 30/,mol-L-1 ) ; nitric oxide (NO), tumor necrosis factor-α(TNF-α), interleukin-1β (IL- 1β) were de- tected. Inducible nitric oxide synthase (iNOS), TNF-α, IL-1β mRNA also were investigated by RT-PCR. Toll like receptor 4 (TLR4) was detected by Western blot. RESULTS Quercetin significantly decreased the NO, TNF-α, IL-1β production, as well as inhibited iNOS, TNF-a, IL-α mRNA syntheses in high-glucose activated microglia cells. Quercetin inhibited the ex- pression of TLR4 at the same times. CONCLUSION Quercetin inhibited high-glucose induced microglia cells activation and might be a useful drug for neuroinflammation relative diseases.
