摘要
目的:研究不同温度条件下副溶血性弧菌的亚致死热损伤程度,最适宜的复苏条件以及复苏后细菌的生长特性。方法:运用传统的固体平板法对不同热处理条件下副溶血性弧菌VpBJ1.1997细胞的亚致死损伤率进行检测,然后通过液体法绘制细菌复苏生长曲线,并结合不同复苏时间下通过扫描电镜观察副溶血性弧菌的形态特征和复苏后亚致死损伤率的测定,最终确定准确的亚致死损伤和复苏条件。结果:副溶血性弧菌在45℃条件下处理时间10min,亚致死损伤率达到最高;35℃复苏3h后,损伤程度最低。结论:不同温度下的亚致死损伤率说明,副溶血性弧菌在30℃条件下培养可以降低亚致死损伤导致的漏检率。
Objective: To study the degree of sublethal thermal injury in Vibrio parahaemolyticus, the optimum resuscitationtemperature of the sublethally injured strains and the growth characteristics of resuscitative strains. Methods: Firstly,we detected the degress of sublethal injury in Vibrio parahaemolyticus BJ1.1997 at different temperatures by usingtraditional solid agar methods. Then, resuscitative growth curves of sublethally injured Vibrio parahaemolyticus BJ1.1997were established by using liquid cultivation method. The morphological characteristics of the sublethally injuries andthe resuscitative strains were observed by using SEM and the optimal conditions of sublethal injury and resuscitationtemperature of Vibrio parahaemolyticus BJ1.1997 were determined. Results: The highest extent of sublethal injury in Vibrioparahaemolyticus BJ1.1997 was obtained after 10 min treatment at 45 ℃ as well as the lowest injury in strains resuscitatedat 35 ℃ for 3 h. Conclusion: According to the measurement of sublethal injury at different temperatures, the probability ofVibrio parahaemolyticus false-negative rate at 30 ℃ can be reduced.
