永磁磁场对大鼠胰岛细胞的增殖、活性影响的实验研究 被引量:2

摘要 [目的]研究在正常和缺氧培养条件下,不同强度永磁磁场对大鼠胰岛细胞增殖及活性的影响。[方法]根据大鼠胰岛细胞生长曲线的情况,选择培养第3天的细胞进行实验。在正常和缺氧培养条件下,采用3.4、5.6、13.8、27.8、52.6、107.0、178.9 mT强度磁场对加磁A-G组进行干预,对照组的细胞不加任何磁场干预,于磁场干预72小时后,利用噻唑蓝(MTT)方法检测各组细胞活性;采用44.8、90.6、182.1mT对加磁a-c组进行干预,对照组的细胞不加任何磁场干预,在磁场干预72小时之后,利用流式细胞仪检测各组细胞增殖情况。[结果]胰岛细胞生长周期吸光度(A)值从第2天(A值0.067±0.021)到第5天(A值0.449±0.113)呈直线生长趋势,故选择培养第3天的细胞进行磁场干预实验。在正常培养条件下,加磁组(A-G组)的MTT值与对照组相比较,差异无统计学意义(P>0.05);在缺氧培养条件下,加磁A组MTT值高于对照组(P<0.01)。在正常培养条件下,加磁组(a、b、c组)的细胞增殖情况随着磁场强度的增加先降低后增加;在缺氧培养条件下加磁组细胞增殖情况随着磁场强度增加而提高。[结论]在正常培养条件下,不同强度的永磁磁场对大鼠胰岛细胞的活性无影响,在高强度的磁场作用下可以提高大鼠胰岛细胞的增殖能力;在缺氧培养条件下,低强度的磁场可提高大鼠胰岛细胞的活性,高强度的磁场则可提高大鼠胰岛细胞的增殖能力。 [Objective] To explore the impact of magnetic field of different intensities on the proliferation and activity in rat islet cells un- der the normal and hypoxic culture conditions. [Methods] According to the islet cells growth curve case, we selected cells cultured 3 days for the experiment. Under the normal and hypoxic culture conditions, using 3.4,5.6,13.8,27.8,52.6,107.0,178.9 mT magnetic field intervene to magnetic A-G group. Control cells received no magnetic field exposure. We used MTT assay to detect cells activity for each group after 72 hours. Using 44.8,90.6,182.1 mT magnetic field intervene to magnetic a-c group. Control cells received no magnetic field exposure. Using Flow Cytometry detect cells proliferation for each group after 72 hours. [Results] The cultured cells grew linearly with optical density (OD) of (0.067±0.021) after 2 days and (0.449±0.113) after 5 days. A-G groups of MTT compared with the control group under the normal culture conditions, and it was no statistically significant (P〉0.05). Under the hypoxic culture conditions, magnetic MTT values higher in group A (P〈0.01). Under the normal culture conditions, cells proliferation increased as the magnetic field strength decreasing first and then increasing in group a-c. And under the hypoxic culture conditions, cells proliferation increased with magnetic field strength. [Conclusion] Under the normal culture conditions, it has no effect on the activity of rat pancreatic islet cells under different intensity permanent magnetic field and it can improve islet cells proliferation on high-intensity magnetic field. Under the hypoxic culture conditions, low-intensity magnetic field can increase the activity of rat pancreatic islet cells, high-intensity magnetic field can increase islet cells proliferation.
