摘要
目的 检测p62/SQSTM1在系统性红斑狼疮(SLE)中的表达情况,明确自噬相关基因p62/SQSTM1与SLE发病的关系.方法 应用实时定量聚合酶链反应技术检测p62/SQSTM1 mRNA在136例SLE患者和25名健康对照组的表达水平,应用蛋白印迹法检测SLE患者中p62/SQSTM1蛋白表达水平.2组计量资料比较采用t检验,计数资料比较采用x2检验.结果 p62/SQSTM1 mRNA在136例SLE患者表达上调,其△Ct值为4.3±1.4,低于健康对照组的5.0±1.8(t=-1.990 6,P=0.048)(△Ct值越小,表达量越大).SLE患者中p62/SQSTM1蛋白质表达水平显著高于健康对照组(0.070 ±0.008和0.046 ±0.007,t=5.943,P=0.001).结论 自噬相关基因p62/SQSTM1表达上调促进SLE的发生.
Objective To investigate the protein and mRNA levels of p62/SQSTM1 protein in systemic lupus erythamtosus (SLE),and explore the relationship between autophage related gene p62/SQSTM1 and the pathogenesis of SLE.Methods In this study,we employed 136 SLE patients and 25 healthy controls.Realtime polymerase chain reaction (PCR) and immunoblotting were adopted to analyze the protein and mRNA levels of p62/SQSTM1 in SLE and healthy populations.All measurement data were expressed as x±s and were tested by t test,and the counting data was analyzed by using x2 test.Results The mRNA levels of p62/SQSTM1 in SLE patients were obviously up-regulated compared to healthy controls (4.3±1.4 vs 5.0±1.8,t=-1.990 6,P=0.048) (the mRNA levels of p62/SQSTM1 were expressed with △Ct values,the value was declined,and the quantity of expression was increased).The expression of p62/SQSTM1 protein levels were significantly up-regulated in SLE compared to those in healthy controls (0.070±0.008 vs 0.046±0007,t=5.943,P=0.001).Conclusion Up-regualted p62/SQSTM1 may promote the occurrence of SLE.
