摘要
目的:探讨完全去前列腺神经后前列腺组织中碱性成纤维细胞生长因子(b-FGF)的表达对前列腺增生的影响。方法:将130只成年SD雄性大鼠随机分成四组,对照组10只,实验组120只,分别用0.01%戊巴比妥钠(1ml/kg)腹腔注射麻醉,实验组破坏支配前列腺的神经后缝合饲养,分别于术后1周,2周,3周各处死40只,完整取出前列腺,分别用于电镜与免疫组化检测。结果:实验组前列腺体积明显缩小,组织轻度水肿,无炎细胞浸润,HE染色与电镜下观察神经纤维显著减少,甚至消失,平滑肌逐渐变薄,腺细胞出现空泡变性,部分细胞器破坏,基底膜萎缩,胶原纤维轻度增生;b-FGF在各组均有表达,实验组随时间延长,表达逐渐减弱,与对照组比较差异有统计学意义(P<0.05);术后1周与2周表达的差异无统计学意义,术后1、2周分别与3周比较,b-FGF表达的差异有统计学意义(P<0.05)。结论:大鼠前列腺去神经后其体积缩小,前列腺组织中b-FGF的表达显著减弱。
Objective: To investigate how b-FGF of prostate tissue affects prostatic hyperplasia after denerva-tion. Method: One hundred and thirty adult male SD rats were randomly divided into four groups, the control group (n=10), the operation group (n1= 40, n2 = 40, n3 = 40). All the experimental rats were anesthetized with 0.01% sodium pentobarbital (1 ml/kg), then their nerves of prostate were dissected. Rats were still alive after the operation. After 1 week, 2 weeks, and 3 weeks respectively the samples of their prostates were obtained. Im- munohistochemistry and electron microscope were used for detection. Result: In the operation groups the size of prostate was significantly reduced with slight edema and without infiltration of inflammatory cells. HE staining technique and electron microscope showed that the number of nerve fibers was significantly reduced and even dis-appeared. Smooth muscles gradually became thin. Glandular cells gradually underwent vacuolar degeneration and the basement membranes became atrophy. Collagen fibers showed a little proliferative. The marker b-FGF ex- pressed in each groups. The level of expression of b-FGF decreased gradually along with time lengthening in the operation groups. There was a statistical difference between the operation subgroups and the control group (P〈 0.05). But there was no difference between n1 and n2, while there existed a statistical difference between nl and n3, n2 and n3 respectively (P〈0.05). Conclusion: The size of prostate and also the expression of b-FGF decrease after denervation.
