摘要
目的探讨新辅助化疗前、后宫颈癌组织OPN mRNA与HIF-1αmRNA的表达变化,分析其作为宫颈癌新辅助化疗敏感度预测因子的可能性。方法采用实时荧光定量PCR法检测20例宫颈癌患者新辅助化疗前、后组织中OPN mRNA与HIF-1αmRNA的表达水平。结果宫颈癌患者新辅助化疗后组织中OPN mRNA与HIF-1αmRNA的表达水平明显下降(P<0.05)。新辅助化疗有效组化疗前、后组织中OPN mRNA与HIF-1αmRNA表达水平均明显低于无效组(P=0.016,P=0.001)。结论宫颈癌患者组织中OPN mRNA与HIF-1αmRNA的表达水平与化疗的敏感性及临床疗效关系密切,可作为宫颈癌新辅助化疗敏感度的重要预测因子。
Objective To investigate the mRNA expression and significance of OPN and HIF-la pre- and post-neoadjuvant chemotherapy in cervical carcinoma, and analyze its possibility to be the predictors for neoadjuvant chemotherapy sensitivity. Methods Real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR) method was used to detect the mRNA levels of OPN and HIF-lα in pre- and post- neoadjuvant chemotherapy in 20 cases of cervical carcinoma tissues. Results The mRNA levels of OPN and HIF-la were significantly decreased after neoadjuvant chemotherapy in patients with cervical cancer tissues (P〈0.05). mRNA levels of OPN and HIF-la in effective chemotherapy group were lower than ineffective group pre- and post-neoadjuvant chemotherapy(P=0.016, P=0.001). Conclusion OPN and HIF-Ia mRNA levels were significantly associated with neoadjuvant chemotherapy chemosensitivity and they might be used as predictors for neoadjuvant chemotherapy sensitivity in cervical carcinoma.
