摘要
目的探讨sCD40L对人脐静脉内皮细胞(HUVEC)分泌TNF-α的影响。方法分组培养HUVEC,应用不同浓度(0、10、20、30、40μg/L)、不同时间(0、10、20、30、40 min)人重组CD40L(r-CD40L)刺激人脐静脉内皮细胞,采用酶联免疫吸附法(ELLISA)测定细胞培养上清液中TNF-α的浓度。结果在相同浓度r-CD40L(20μg/L)刺激下,HUVEC在20 min分泌TNF-α水平最高,5组间比较差异有统计学意义(F=500.15,P<0.01);随r-CD40L刺激浓度增加,HUVEC分泌TNF-α水平增高,呈浓度依赖性,5组间比较差异有统计学意义(F=128.75,P<0.01)。结论 HUVEC可在r-CD40L刺激的早期分泌TNF-α达峰值,r-CD40L刺激HUVEC分泌TNF-α呈浓度依赖性。
Objective To investigate the effect of sCD 40 L on the secretion of tumor necrosis factor-α( TNF-α) in human umbilical vein endothelial cells ( HUVEC ) .Methods The HUVECs were cultured in different groups,and HUVECs were stimulated by r-CD40L at different concentrations (0,10,20,30,40 μg/L) and in different time points (0,10,20,30,40 min),then the concentrations of TNF-αin supernatant were detected by enzyme-linked immunosorbent assay (ELISA ) .Results The levels of TNF-αsecreted by HUVEC in supernatant were the highest at 20 minutes after stimulation by r-CD40L (20 μg/L), there were significant differences among the 5 different time groups ( P 〈0.01).With the increase of concentration of r-CD40L,the levels TNF-αsecreted by HUVEC in supernatant were increased in a dose -dependent manner , there were significant differences among the 5 different dose groups ( P 〈0.01).Conclusion The levels of TNF-αsecreted by HUVEC may reach peak at early time after r-CD40L stimulation, moreover, which is in a concentration -dependent manner .
