摘要
目的探讨脂多糖(lipopolysaccharide,LPS)所致大鼠急性肺损伤时环氧合酶(COX)-2抑制剂塞来昔布对外周血趋化因子白细胞介素(interlukine,IL)-8和单核细胞趋化蛋白(monocyte chemoattractant protein,MCP)-1表达的影响。方法 45只大鼠随机分为3组:对照组(n=15),给予等量的0.9%氯化钠溶液;LPS组(n=15):尾静脉注射LPS(5 mg/kg)复制急性肺损伤模型;塞来昔布治疗组(n=15):注射LPS复制急性肺损伤模型后30 min塞来昔布灌胃(20 mg/kg),于3 h后放血处死动物。采用酶联免疫吸附试验(ELISA)检测3组大鼠外周血IL-8和MCP-1的表达水平。结果 LPS组和对照组相比外周血IL-8和MCP-1的表达差异均有统计学意义(P<0.05);塞来昔布组和LPS组相比,差异有统计学意义(P<0.05)。结论塞来昔布可能通过抑制趋化因子IL-8和MCP-1的表达,改善大鼠急性肺损伤反应程度。
Objective To investigate the effects of celecoxib on the expressions of peripheral blood chemokines-interlukine 8 ( IL-8) and monocyte chemoattractant protein ( MCP-1 ) in rats with acute lung injury induced by lipopolysaccharide ( LPS) .Methods Forty-five rats were randomly divided into three groups:control group,LPS group and celecoxib treatment group ,with 15 rats in each group.The rats in control group were given 0.9%sodium chloride solution ,the animal models with acute lung injury were established by intravenous injection LPS(5 mg/kg) and the rats in celecoxib treatment group were given celecoxib (20mg /kg) at 30min after LPS induction,then the rats were sacrificed after 3h.The expression levels of IL-8 and MCP-1 in peripheral blood of rats were detected by ELISA .Results There were significant differences in the expression levels of IL-8 and MCP-1 between LPS group and control group ( P 〈0.05).There were significant differences in the expression levels of IL-8 and MCP-1 between celecoxib treatment group and LPS group ( P 〈0.05).Conclusion Celecoxib can improve the reaction extent of rats to acute lung injury by inhibiting the expression levels of IL -8 and MCP-1.
