血府逐瘀汤含药血清体外诱导MSC形态学改变及表达cTnT的实验研究

摘要 目的:探讨复方血府逐瘀汤含药血清体外诱导MSC分化为心肌细胞的能力,作为细胞移植治疗冠心病、心肌梗死等缺血性心脏病的种子细胞,旨在提高细胞移植治疗冠心病的安全性。方法:分离培养Wistar大鼠骨髓基质干细胞(MSC),用血府逐瘀汤含药血清诱导后,倒置荧光相差显微镜观察细胞形态学的改变,免疫细胞荧光染色法检测cTnT的表达。结果:MSC经过血府逐瘀汤含药血清体外诱导后,MSC的细胞形态发生了变化,具有和5-氮胞苷类似的诱导作用,朝着心肌细胞形态学的方向发展,心肌肌钙蛋白T出现了阳性表达。结论:复方血府逐瘀汤含药血清体外可以诱导大鼠MSC分化为心肌样细胞。 Objective : To investigate the ability of blood serum of Xue-fu Zhu-yu Decoction on inducing MSC differentiating into myocardial ceils in vitro, which could be the seed ceils in cellular transplantation to treat ische- mic heart diseases such as coronary heart disease and myocardial infarction in order to improve the safety of cell transplantation in the treatment of coronary heart disease. Methods : The bone marrow stromal stem cells (MSC) of Wistar rat were isolated and cultured, and induced by blood serum of Xue-fu Zhu-yu Decoction. The cell morphol- ogy changes were observed with inverted fluorescence microscope, and immunofluorescence staining was used to detect the expression of cTnT. Results : After induced by blood serum of Xue-fu Zhu-yu Decoction in vitro, MSC changed in the cell morphology, and was similar to the inducing function of 5- azacytidine. Then MSC developed toward the myocardial cell morphology, and cardiac troponin T showed positive expression. Conclusion: Blood ser- um of Xue-fu Zhu-yu Decoction can induce rat MSC to differentiate into cardio-myoid cells in vitro.
