摘要
目的探讨黄芪皂甙Ⅳ联合脂肪源性干细胞(ADSC)对大鼠坐骨神经缺损后运动功能恢复的影响。方法将40只清洁级雄性SD大鼠随机分为4组各10只,4组均采用化学萃取脱细胞方法制备大鼠缺损脱细胞神经支架(ARSN)。ARSN组注入等剂量DMEM于ARSN;黄芪皂甙Ⅳ组注入等剂量DMEM于ARSN,并于术前5 min及术后12、24 h腹腔注射黄芪皂甙Ⅳ(20 mg/kg);ADSC组注入等剂量ADSC(1×106/mL)于ARSN;黄芪皂甙Ⅳ+ADSC组同时采用黄芪皂甙Ⅳ组、ADSC组的方法。用各组的神经移植体桥接长1 cm坐骨神经两断端间的缺损造模,术后8周取材。检测各组坐骨神经功能指数(SFI)、神经电生理参数(神经传导速度、潜伏期、波幅)及胫前肌湿重比率。结果与ARSN组比较,黄芪皂甙Ⅳ组、ADSC组和黄芪皂甙Ⅳ+ADSC组术后各时点SFI增高,神经传导速度增快,潜伏期缩短,波幅增大,胫前肌湿重比率增高(P均<0.05);与黄芪皂甙Ⅳ组比较,黄芪皂甙Ⅳ+ADSC组术后56 d SFI增高,神经传导速度增快,潜伏期缩短,波幅增大,胫前肌湿重比率增高(P均<0.05)。结论黄芪皂甙Ⅳ联合ADSC移植能促进大鼠神经再生和运动功能恢复,其作用优于单独应用黄芪皂甙Ⅳ或ADSC。
Objective To investigate the effects of astragaloside Ⅳ combined with adipose tissue-derived stem cells (ADSC) on motor functional recovery after acellular nerve allograft (ARSN) repair of the sciatic nerve defects in rats.Methods Forty SD rats were randomly divided into 4 groups,10 in each group,and 4 groups were treated by chemical extraction method for the preparation of rat defect ARSN.In the ARSN group,the same dose of DMEM was injected into ARSN; in the astragaloside Ⅳ group,the same dose of DMEM was injected into ARSN,moreover,astragaloside Ⅳ (20 mg/kg) was intraperitoneally injected into ARSN 5 min before and 12 h,24 h after the operation; in the ADSC group,the same dose of ADSC (1 × 10^6/mL) was injected into ARSN; in the astragaloside Ⅳ + ADSC group,both methods used in the astragaloside Ⅳ group and ADSC group were applied here.The nerve graft from each group was interposed into sciatic nerve gap.At 8 w after operation,the materials were obtained.The sciatic function index (SFI),electrophysiology (conduction velocity,latent period and wave amplitude) and the ratio of tibialis anterior muscle wet weight were tested.Results Compared with the ARSN group,SFI at each time point after operation,nerve conduction velocity,wave amplitude,ratio of tibialis anterior muscle wet weight were significantly increased,and latent period was decreased in the astragaloside Ⅳ group,ADSC group and astragaloside Ⅳ + ADSC group (all P 〈 0.05).Compared with the astragaloside Ⅳ group,56 d SFI,nerve conduction velocity,wave amplitude and the ratio of anterior tibialis muscle wet weight were increased,and latent period was decreased in the astragaloside Ⅳ + ADSC group (all P 〈 0.05).Conclusion The astragaloside Ⅳ combined with ADSC transplantation enhances the nerve regeneration and promotes motor functional recovery,whose effect is better than either of the single treatment.
