摘要
目的观察预先静脉注射葡萄糖酸钙溶液,对间斑寇蛛毒致痛大鼠行为学的影响;评估葡萄糖酸钙溶液对抑制间斑寇蛛毒致痛作用的疗效。方法将雄性SD大鼠80只随机分为葡萄糖酸钙治疗组与致痛组,治疗组大鼠预先尾部静脉注射葡萄糖酸钙溶液,两组大鼠随机分为5组,分别以低剂量(10μL/kg)、中低剂量(40μL/kg)、中剂量(100μL/kg)、高剂量(300μL/kg)的间斑寇蛛毒素及福尔马林(10%甲醛溶液)进行足底注射,观察记录两组大鼠注射后2、15、30min及2h缩足、舔足次数。结果 (1)中低剂量组和高剂量组的蛛毒致痛大鼠缩足次数在15min内均高于福尔马林致痛组,然而30min却低于福尔马林致痛组(P<0.05);(2)15min时致痛组及治疗组大鼠缩足次数均与毒素浓度呈显著的正相关(r2=0.597,P<0.05;r2=0.426,P<0.05);(3)葡萄糖酸钙治疗组与各毒素致痛组相比,大鼠缩足次数均减少(P<0.05)。结论间斑寇蛛毒素致痛大鼠的作用相对于福尔马林在30min内更为强烈,但是30min后则较福尔马林组有所减轻,预先静脉注射葡萄糖酸钙的方法对间斑寇蛛毒致痛有明显的镇痛作用。
Objective This study was designed to observe behaviors of rats which were given calcium gluco- nate intravenously after the rats were poisoned by the toxic components from Latrodectus tredecimguttatus spiders and the analgesia function of calcium gluconate was evaluated. Methods 80 male SD rats which had been injected with calcium gluconate beforehand were randomly divided into five groups, and then they were given spider toxin and formalin with low dose (10 μL/kg), sub-medium dose (40 μL/kg), medium dose (100 μL/kg), and high dose (300 μL/kg) respectively. The nociceptive response and flinching times were observed and recorded. Results (1) The flinching times were more frequent in sub-medium dose group and high dose group than those in formalin group in 15 minutes, however, less frequent than those in formalin group in 30 minutes (P 〈0.05). (2) The fliching times caused by pain had significant positive correlation with concentration of spider toxin (r2 = 0. 597, P 〈0.05, r2 = 0. 426, P 〈0.05). (3) The flinching times were reduced in the calcium gluconate treatment group (P 〈0.05). Conclusion The pain caused by Latrodectus tredecimguttatus spiders toxin was more intense than that caused by formalin in 30 minutes. However, it was relieved after 30 minutes. Therefore, the pain caused by Latrodectus tredecimguttatus spiders toxin could be relieved significantly when the calcium gluconate were given beforehand.
