摘要
目的探索实验性矽肺大鼠肺组织中白细胞介素-17A(IL-17A)的表达,为开展IL-17A在矽肺纤维化的研究提供依据。方法16只健康雄性SD大鼠随机分为两组,矽肺组和对照组各8只,矽肺组大鼠气管非暴露注入粉尘50mg,对照组大鼠注入生理盐水。染尘40d处死大鼠,取肺组织进行病理学观察,实时荧光定量PCR(Realtime qPCR)法定量检测肺组织中IL-17A mRNA的转录水平,免疫组化法检测两组大鼠肺组织中IL-17A的蛋白表达水平。结果矽肺组大鼠肺组织中IL-17A mRNA与蛋白表达水平明显高于对照组(P<0.05)。结论IL-17A在矽肺大鼠肺组织中高表达,其可能参与了矽肺中晚期肺纤维化过程。
Objective Exploring the expression of IL-17A and studying it's role in the pathogenesis of silicosis pulmonary fibrosis. Methods Sixteen healthy male SD rats were randomly divided into a silica exposure group and a control group equally. Then 1.0 ml silica suspension (50 g/ml) was administrated via intra-tracheal instillation once while equal amount of saline was instilled to the rats of the control group. All rats were sacrificed on the 40th day after instillation and samples of lung tissue were collected. The pathological changes were determined by HE staining and the expression of IL-17A was investigated by immunohistochemistry and real-time quantitative polymerase chain reaction ( Realtime-qPCR ). Results Compared with the control group, the mRNA and protein expression of IL-17A both enhanced and had significant different in the lung of the silicotic rats(P〈0.05). Conclusions The high expression of IL- 17A in the lung of the silicotic rats indicated that IL-17A may he involved in the pathological process of silicosis pulmonary fibrosis in later stage of silicosis.
