摘要
文本主要探讨叶下珠提取物对人肝癌细胞的抑制作用及其对Bcl-2的表达的影响。在倒置显微镜下利用利用MTT比色法观察体外培养的人肝癌细胞株(BEL-7404)在不同浓度的叶下珠提取物作用后的形态变化,通过这种方法来确定各种浓度下的叶下珠提取物对于肝癌细胞的抑制程度。同时制作爬片,利用细胞免疫化学法测定Bcl-2在细胞中的表达情况。结果显示叶下珠提取物对于肝癌细胞有抑制作用,且抑制效果在实验范围内随着药物的浓度增加而增强。并且,叶下珠提取物能够减弱Bcl-2在体内的表达程度。说明叶下珠提取物通过下调细胞凋亡抑制基因Bcl-2的表达,从而促进肝癌细胞的凋亡。
The study mainly discusses the effects of Phyllanthus extract on human hepatoma cell line and its on the expression of Bcl-2. Under an inverted microscope observation using the MTT assay cultured human hepatoma cell line (BEL-7404) in different concentrations of Phyllanthus extracts morphological changes after the action, through this method to determine the concentrations of various leaf Phyllanthus extract the degree of inhibition of human hepatoma cells. While cells grow on cover slips production, determined using cell immunochemical Bcl-2 expression in ceils. The results showed that Phyllanthus extracts inhibited the bepatoma ceils, and the inhibitory effect within the experimental range increases with the concentration of the drug increased. And, Phyllanthus extracts can decrease Bcl-2 expression levels in vivo. Description Phyllanthus extract inhibited apoptosis by down-regulating gene expression of Bcl-2, thereby promoting apoptosis of hepatoma cells.
