摘要
用蛋白酶K消化法提取萧氏松茎象总DNA,并对细胞色素C氧化酶亚基Ⅰ(cytochrome c oxidaseⅠgene,COⅠ)序列分析发现,COⅠ序列4种核苷酸量平均为A=30.2%,C=17.3%,G=15.4%,T=37.0%。将所获得的5个不同地区萧氏松茎象个体COⅠ序列排序后,比较形成了一个含1 209个位点的矩阵,其中1 174个不变位点,35个变异位点,所占比例为2.89%,序列的A+T含量较高。遗传距离、核酸和氨基酸变异及系统发育树分析表明:萧氏松茎象与树皮象属亲缘关系最近,且分子水平上萧氏松茎象与树皮象属之间较之树皮象属内种间无明显差异,对目前的分类地位提出质疑。
The total genomic DNA of Hylobitelus xiaoi Zhang was extracted by means of digestion with proteinase K digesting,sequence CO Ⅰ.The result showed,the average content of A was 30.2%,that of C was 17.3%,that of G was 15.4%,that of T was 37.0% in sequences of CO Ⅰ.After alignment the size of sequences from five individuals of different sampling areas was 1 209 bp,including 35 mutations (the mutation rate was 2.89%),which showed the higher contents of A + T.The analysis of genetic distance,variation ratio of nucleotide and amino acid,and the phylogenetic tree showed that Hylobitelus xiaoi is closely related to the species of Hylobius.It is proved that there is no obviously difference between Hylobitelus xiaoi and Hylobius sp.At molecular level,its present taxonomic status of Hylobitelus xiaoi is doubtful.
