摘要
分别以蒴果(种子)、花茎和地下茎为外植体对白芨进行组织培养和快速繁殖。结果表明:种子播种于培养基MS+KT 1.0 mg/L+NAA 0.5 mg/L进行白芨原球茎的诱导和芽的分化效果最好,随后转接到培养基MS+6-BA 0.45 mg/L+NAA 0.4 mg/L进行壮苗生根培养,30 d后将生根无菌苗移栽到蛭石中,成活率可达到96%。本文采用无菌播种和组织培养方法获得无菌苗,并进行壮苗生根培养与移栽,不仅繁殖成本低、培养周期短,且操作简单易行便于推广,可为其优良品种的开发利用和快速繁殖提供技术支持。
Capsule (seed),floral axises and underground corms of Bletilla striata were used as the explants for the tissue culture and rapid propagation.It was showed that:the seeds germinated on MS + KT 1.0 mg/L + NAA 0.5 mg/L was the best for protocorm induction and bud differentiation,and those transferred to the vermiculite for lustihood and rooting culture on MS +6-BA 0.45 mg/L + NAA 0.4 mg/L for 30 d had a survival rate of 96%.The sterile seedlings of Bletilla striata were obtained by asepsis sowing and tissue culture,and then subjected to lustihood and rooting culture and transplanting,which is not only of low reproduction cost,short culture period,but also simple operation and easy promotion.Therefore,it can provide technical support for the development,utilization and rapid propagation of fine varieties of Bletilla striata.
