TNF-α对损伤的内侧副韧带和前交叉韧带中BMP-1基因表达的影响

摘要 目的:就TNF—α对损伤的内侧副韧带和前交叉韧带中BMP-1基因表达的影响进行探讨。方法:选择重庆医科大学附属第一医院骨科教研室在2011年09-11月时间内所收集到的4例滑膜组织和交叉韧带标本,其中女2例,男2例,截肢膝关节相应组织是在无菌条件下用镊子和剪刀来取得。先细胞培养,其次,pET32a(+)-Tat—PTD—VP3重组质粒构建示,分别测定半定量PCR、实时定量PCR。结果:本组资料研究表明,TNF-α作用时间分别为24h、12h、6h、2h,作用浓度为5ng/ml。对于那些已经损伤的MCL细胞中,BMP-1的表迭随着TNF-α浓度的不断增加也会出现避步的下降趋势,用实时定量PCR来进行显示,BMP-1表这为117%、70%、62%、41%。而对于那些已经损伤的ACL细胞中,BMP-1的表达随着TNF-α浓度的不断增加会出现逐渐下降的趋势。用实时定量PCR来进行显示,BMP-1表达从最初的69%,下降到最后的27%。结论:TNF-α能够有效地下调ACL作用,以及前交叉韧带(ACL)和内侧刮韧带(MCL)中BMP~1基因表述都出现了较大的下降,本实验为有效促进ACL自我修复,早期抑制TNF-α浓度提供了较好的理论依据。 Objective:To explore effects of BMP-1 TNF- on the injury of the medial collateral ligament and anterior cruciate ligament in gene expres- sion. Methods:4 cases of synovial tissue Department of orthopedics,First Affiliated Hospital of Chongqing Medical University collected in 2011:09 - 11 cruci- ate ligament specimens,including 2 cases of female,male 2 cases, amputation of knee joint corresponding tissue is achieved with tweezers and scissors under sterile conditions. Second, cell culture, pET32a ( + ) to construct the recombinant plasmid of - Tat- PTD- VP3, semi- quantitative PCR, real- time quanti- tative PCR were measured. Results:the study shows that, TNF-α action time were 24h, 12h, 6h, 2h, concentration is 5ng' ml. For those who have damage in MCL cells,the expression of BMP-1 with increasing TNF- concentration will gradually decline, real-time quantitative PCR was used to display,BMP-1 expression was 117 %, 70 %,62 % ,41%. And to all those who have injured ACL ceils, the expression of BMP-1 with increasing TNF- concentration will gradually decline. Real--time quantitative PCR was used to show the expression of BMP-1, from the initial 69 %, down to the last 27 %.
