哈维氏弧菌BB170冷冻干燥保护剂的选择

摘要 哈维氏弧菌BB170是目前公认的检测细菌发酵液中二类信号分子即AI-2信号分子的指示菌,其产生的荧光强度强弱对试验成功有至关重要的作用。为了保证在真空冷冻干燥后其产生的荧光强度仍能达到最佳状态,首先通过单因素试验,以荧光强度为指标,筛选出对哈维氏弧菌BB170效果明显的保护剂为脱脂乳、L-谷氨酸钠、蔗糖和葡萄糖,再通过4因素3水平正交试验,确定其影响效果为脱脂乳>L-谷氨酸钠>蔗糖>葡萄糖,最佳保护剂配方为:10%脱脂乳、1%L-谷氨酸钠、2%蔗糖和1.25%葡萄糖。在常温下存放2个月后达到稳定,且能满足AI-2信号分子的检测。 Vibrio harveyi BB170 is used as indicator bacteria to detecte autoinducer-2 (I. e. AI-2) signal molecular of bacterial fermentation and its fluorescence intensity is crucial for the successful test. In order to ensure the fluorescence intensity still can reach the best state after vacuum freeze-drying, we found that skim milk, L-sodium glutamate, sucrose and glucose were obvious protective agent for Vibrio harveyi BB170 as the target of fluorescence intensity through the single factor experiment. By four factors and three levels orthogonal experiment, results showed its effect was skim milk〉L-sodium glutamate 〉sucrose〉glucose. The best protective agents formula was 10% skim milk, 1% L-sodium glutamate ultimately, 2% sucrose and 1.25% glucose. Bacterial powder was stable and can meet the detection of AI-2 signal molecules after two months preservation at room temperature.
