高效阴离子交换色谱—脉冲安培检测法分析贼小豆中的糖类化合物

Analyze carbohydrate compounds in Vigna minima (Roxb.) with high performance anion-exchange chromatography using pulsed amperometric detection

摘要 建立了高效阴离子交换色谱法同时测定贼小豆中9种糖类化合物含量的分析方法。用CarboPacTMPA10阴离子交换分离柱和脉冲安培检测器,以0.25mol/L氢氧化钠和水等度洗脱,流速0.80mL/min,柱温30℃。结果表明:25rain内能完成9种糖的分离,在0.05—25.0mg/L范围内呈良好的线性关系,相关系数大于0.9993,检出限为0.05--1.20μg/L,加标回收率为91.66%~103.2%,相对标准偏差RSDd、于2.3%(n=6)。方法灵敏度高,前处理简单快速、选择性好、无需衍生,用于郑州地区野生贼小豆中糖类化合物的快速检测,结果较满意。 A chromatographic method was developed for the simultaneous determination of 9kinds carbohydrate compounds in Vigna minima(Roxb.) using high performance anion-exchange chromatography coupled with pulsed amperometric detection(HPAEC-PAD). The analyzes were separated on CarboPacTM PAl0 anion exchange separation column using 0.25 mol/L NaOH solution and water as mobile phase in the isocratic elution at flow rate of 0.80 mL/min and detected with column temperature 30 ~C. The results showed that: nine carbohydrate compounds can be separated well within 25 min. Linearity ranges for all the 9 kinds carbohydrate compounds were 0.05~25.0 mg/L with a correlation coefficient exceeding 0.9993. Under the optimum conditions, the limits detection (3S/N) were 0.05~1.2 IJg/ L (25 IJL injection, and the quantitative analysis was based on the peak areas) respectively. Moreover, the standard addition recoveries were 91.66%~103.2%, and the relative standard deviations(RSD) less than 2.3%(n=6). This method is simple, rapid, high sensitive, and has good selectivity, and needn't derivation in the process of determination. It is quite suitable for the determination of carbohydrate compounds in Vigna minima(Roxb.) in Zhengzhou area with satisfying results.
