抗包虫药物的体外药效对比实验研究

摘要 目的 :包虫病是一种危害性很大的人畜共患病 ,除了甲苯咪唑 (MBZ)和阿苯达唑 (ABZ)这两种公认的有效药物以外 ,目前尚无新的更有效的药物出现。本实验通过体外试验进行了氧苯达唑 (OBZ)、阿苯达唑和阿苯达唑脂质体 (L - ABZ)的体外抗细粒棘球蚴原头节作用的比较 ,探讨氧苯达唑在抗包虫病方面的作用 ,为探索新的抗包虫病药物奠定基础。方法 :将氧苯达唑、阿苯达唑和阿苯达唑脂质体分别加入 RPMI16 40培养液中 ,配成高 (2 5 0 0 ng/ml)、中 (5 0 0 ng/ ml)和低 (10 0 ng/ ml) 3个浓度的加药培养液 ,将用无菌方法采集的自然感染的羊细粒棘球蚴原头节 (活头节占 95 %以上 )加入上述加药培养基中进行培养 ,每天计数其死亡率 ,方法如下 :取一滴培养液 ,0 .1%伊红溶液染色 5 min,在普通光镜下观察 ,死亡原头节红染着色 ,伴有结构破坏 ;活头节则拒染不着色 ,计数每一视野内着色 /不着色头节数目 ,以着色者所占观察数之百分比作为原头节的死亡率 ,观察 3个视野 ,计算平均死亡率。每天计数 1次 ,直到对照组的原头节也全部死亡为止。实验过程中不更换培养基。结果 :(1)原头节死亡率氧苯达唑、阿苯达唑、阿苯达唑脂质体与对照组相比均有统计学意义 (P <0 .0 5 ) ,且氧苯达唑、阿苯达唑在低浓度时与对照组相比有统计? Objective: The effect of antihydatidosis of oxibendazole、albendazole and lip albendazole on protoscoleces of echinococcus granulosus was compared in vitro. Methods: Protoscoleces of echinococcus granulosus was cultured in the different concentration(high、middle、low) of oxibendazole、albendazole and lip albendazole in 1640 culture media respectively in vitro. The daily mortality in each group was counted until the day of the protoscoleces of control completely dead. Results: The significant difference of the mortality between the three drugs with various concentrations and the control group was found respectively(P<0.05); however,in lip albendazole group the statistic difference only in high concentrations was found(P<0.05); the antiprotoscoleces effect of these drugs compared each other, no statistic differences was found(P>0.05). Conclusions: The significant antiprotoscoleces effect of the three drugs was found in vitro. Oxibendazole, a new anti hydatid drug,had the similar antiprotoscoleces effect with albendazole in vitro;there was no significant effect on their antiprotoscoleces by the formulation changing between albendazole.and lip albendazole only in vitro.
