摘要
实验旨在研究热应激条件下小鼠体内睾酮浓度的变化以及外源性睾酮对TM4细胞(小鼠支持细胞)中热休克蛋白(HSPs)mRNA表达的影响。将40只7周龄的雄性昆明种小白鼠随机分为4组,每组10只,对照组在(23±2)℃下饲养,实验组置于(39±0.5)℃下热应激处理,观察不同热应激时间(0.5、1 h和2 h)下小鼠体内睾酮浓度的变化;以TM4细胞为实验材料,研究外源性睾酮对HSPs mRNA表达的影响。结果表明:与对照组相比,实验组小鼠血清和睾丸中的睾酮浓度分别在热应激1 h和2 h后显著下降(P<0.05);在热应激处理过的TM4细胞中添加2μmol/L睾酮能够显著增加其Hsp72、Hsp90和Hsp105 mRNA的表达水平(P<0.05),并且Hsp72和Hsp105mRNA的上升可以被雄激素受体抑制剂Flutamide抑制;但是在未经过热应激处理的TM4细胞中睾酮对上述HSPs的表达没有显著影响。结果显示,热应激降低了小鼠体内的睾酮浓度;热应激时外源性睾酮可通过雄激素受体上调Hsp72和Hsp105 mRNA的表达。
The experiment was conducted to evaluate the effects of heat stress on androgen level in mice and exogenous androgen on the expression of HSPs mRNA in the TM4 cells (Sertoli cells of mouse). 7-week-old male Kunming mice were used to observe the changes of androgen level via processing different heat-stress (39±0.5)℃ time (0.5h, 1 h, 2 h). We also evaluated the effects of exogenous testosterone on the expression of HSPs mRNA in the TM4 cells. The results showed that compared with the control groups (24± 1)℃, both serous and testicular testosterone concentrations of the experimental mice decreased significantly after heat stress lh and 2h (P 〈0.05); Addition of 2μmol/L testosterone in culture medium increased the expression of Hsp72, Hsp90 and 105 mRNA significantly in heat- stressed TM4 cells (P〈0.05) while not in cells cultured in normal temperature, this increase of Hsp72 and Hspl05 mRNA can also be inhibited by Flutamide, the androgen receptor inhibitor, however, no significant decrease was detected in Hsp90 mRNA. In conclusion, the level of androgen decreases in mouse under heat stress; Hsp72 and Hspl05 mRNA expression can be raised by exogenous testosterone through AR in heat-stressed TM4 cells.
