VX-680对T24细胞增殖的抑制作用及对CyclinD1和CyclinB1表达的影响 被引量:3

摘要 目的:探讨AuroraA激酶抑制剂VX一680对人膀胱癌T24细胞增殖的抑制作用及对CyclinB1和CyclinB1mRNA表达的影响及意义。方法:体外培养T24细胞.分别给予不同浓度及不同作用时间的VX-680.采用MTT法检测细胞增殖能力:RT—PCR检测T24细胞Cyclin B1和Cyclin B1mRNA的表达水平,结果:MTT结果显示:100~500nmol/LVX-680作用24、72h均对T24细胞有明显抑制作用.呈浓度依赖性且72h时抑制效果更显著:100~500nmol/LVX-680作用72h后,各组T24细胞CyclinB1和CyclinB mRNA的表达水平下调,呈浓度依赖性。结论:Aurora激酶抑制剂VX-680可能通过下调CyclinB1和CyclinB1mRNA表达显著抑制人膀胱癌T24细胞增殖,且具有浓度依赖性。
