摘要
目的 :探讨以 HPL C法对淡豆豉中异黄酮的定量分析。 方法 :采用 HPL C法 ,YWGC1 8柱 ,以甲醇∶水∶乙酸( 10∶ 10∶ 1)为流动相 ,检测波长 2 6 0 nm ,测定淡豆豉中染料木素、大豆黄素含量。 结果 :淡豆豉中游离染料木素含量为( 2 30 .6 4± 9.14)μg/ g、大豆黄素含量为 ( 2 6 4.2 6± 4.2 2 )μg/ g;用盐酸水解处理后样品中染料木素总含量为 ( 2 76 .0 0± 7.81)μg/ g,大豆黄素总含量为 ( 2 87.6 5± 5 .70 ) μg/ g。 结论 :用 HPL C法测定淡豆豉中异黄酮类成分含量 ,建立的方法准确、简便 ,可用于控制和评价淡豆豉的品质。
Objective: To provide evidence and method for evaluating the quality of Seme sojae preparatum (SSP). Methods: Contents of the isoflavones (genistein and daidzein) in SSP was measured by HPLC [ a column of YWGC 18 , methanol∶water∶acetic acid (10∶10∶1) as a mobile phase, detective wave 260 nm]. Results: The content of genistein was (230.64±9.14)μg/g and daidzein was (264.26±4.22) μg/g in SSP. The total contents of genistein and daidzein were (276.00± 7.81) and (287.65±5.70)μg/g respectively after hydrolysis by HCl. Conclusion: Determination of isoflavones in SSP by HPLC is acurate and conveniet. The results show that it is a suitable method for evaluating the quality of SSP. [
