摘要
目的 :探讨联合培养的血管平滑肌细胞 (VSMC)对内皮细胞 (EC)粘附能力的影响。 方法 :应用免疫荧光细胞化学、激光共聚焦扫描显微镜和图像分析等技术 ,以静态条件下单独培养的 EC为对照组 ,观察与 VSMC联合培养的 EC细胞外基质 Fn,L n和 Col 的构筑方式及其含量的变化。结果 :与单独培养的 EC相比 ,联合培养的 EC细胞外基质各组分的构筑有明显的变化 ,且含量也发生变化 ,Fn,L n增多 ,Col 减少。结论 :与 VSMC联合培养的 EC的粘附能力可能增强 ,为改进血管 EC种植的组织工程技术提供了部分实验依据。
Objective: To stady the effect of co cultured vascular smooth muscle cells (VSMCs) on the adhesion ability of endothelial cells (ECs). Methods: Organization and composition of extracellular matrix (ECM) , fibronectin (Fn), laminin (Ln) and collagen type Ⅳ (ColⅣ) by co cultured ECs with VSMCs was investigated with the methods of immuno fluor cytochemistry, laser confocal scanning microscopy and image analysis. The control was ECs cultured alone. Results: In static condition, there was clear alternation in each part of ECM organization between co cultured ECs and ECs cultured alone, and the content of co cultured ECs varied, with an increase of Fn and Ln and a decrease of ColⅣ. Conclusion: The capability of against stress and adhesion of co cultured ECs are strengthened. Our results may also provide some theoretical and experimental foundation for improving tissue engineering of vascular endothelial cells seeding. [
