摘要
目的对AhR、CYP1B1在哮喘模型小鼠肺组织及外周CD4+T细胞中的表达特征进行研究,探讨AhR、CYP1B1在哮喘发病中的作用。方法建立哮喘小鼠模型,制备肺组织石蜡切片进行病理学检查。磁珠阳性分选法从脾脏中分离获得外周CD4+T细胞,利用RT-PCR半定量技术分别测定正常组和哮喘组小鼠肺组织及外周CD4+T细胞中AhR、CYP1B1 mRNA的表达量。结果两组小鼠肺组织和外周CD4+T中均有表达AhR、CYP1B1。哮喘组肺组织及外周CD4+T细胞中AhR、CYP1B1表达均较正常组低(P<0.05)。结论 AhR、CYP1B1在支气管哮喘的发病过程中发挥作用,可能是支气管哮喘治疗的新靶点。
Objective To investigate the expression of AhR,CYP1 B1 in tissue of the lung and the peripheral CD4 T lymphocytes in the mouse asthma model and the role of AhR,CYP1B1 in the pathogenesis of asthma. Methods The mouse asthma model was estab- lished. The paraffin slices of tissue in the lung were made to assess the histological changes by means of histopathology. Bead positive se- lection method is used to get peripheral CD4 +T cells from the spleen. The mRNA expression of AhR,CYP1B1 genes in the lung tissue and the peripheral CD4 +T lymphocytes isolated from the spleen in the asthmatic group and the normal group were detected by reverse tran- scription polymerase chain reaction( RT - PCR). Results The expression of AhR,CYP1 B1 genes were detected in the lung tissue and the peripheral CD4 +T lymphocytes. The expressions of AhR, CYP1B1 decreased signillcantly both in the lung tissue and in the peripheral CD4 + T lymphocytes in the asthmatic group compared with those in the normal group. Conclusion AhR,CYP1 B1 play a role in the path- ogenesis of asthma and maybe a new target of the asthma therapy.
