摘要
目的 :研究瘤体内直接注射脂质体包裹的人 IFNα基因的抗肿瘤效果。 方法 :在鳞癌荷瘤裸鼠的瘤体内注射体外已制备好的用脂质体包裹的 IFNα DNA,观察肿瘤生长状况及小鼠存活情况。结果 :瘤体内注射脂质体包裹的人 IFNα基因后 ,荷瘤裸鼠肿瘤的生长明显受抑 ,其中部分荷瘤裸鼠的肿瘤消失 ;荷瘤裸鼠的存活期明显延长 ,部分荷瘤裸鼠可长期存活。病理结果显示 ,治疗后的肿瘤细胞形状不一 ,有多数细胞质崩解 ,破坏明显。结论 :瘤体内注射脂质体包裹的 IFNα基因后 ,可在瘤体内原位直接转染肿瘤细胞并破坏该肿瘤细胞 ,从而达到抑制肿瘤生长的效果。
Objective: To investigate the antitumor effect of the injection of IFNα DNA liposome in tumor. Methods: BMGNeo IFNα DNA was transfered into the tumor of squamous carcinoma by the liposome method. The tumor growth and survival rate of nude mice were observed. Results: After intratumor injection of IFNα DNA liposome, the tumor growth was dramaticly inhibited and the survival of the nude mice was prolonged significantly, some tumors disapppeared and the nude mice lived well. Pathology showed that the shape of the tumor cells was different and the cytaplasm was damaged. Conclusion: Human IFN α gene may be transfered into squamous carcinoma cells by intratumor injection. The tumor growth can be inhibited and the programmed cell death of oral squamous carcinoma cells may be induced by injecting IFNα DNA liposome intumor. [
