β-arrestins, important soluble proteins mediating receptor desensitization, have diverse biological functions, such as regulating cell proliferation, cell survival, apoptosis and gene transcription. β-arrestins regulate inflammatory and immune reactions by inhibiting the basal activity of pro-inflammatory transcription factor NF-κB and participating in Toll-like receptors (TLR)/NF-κB signal pathway-mediated NF-κB activation. β-arrestins are involved in the pathogenesis of various inflammatory diseases, such as inflammatory bowel disease, rheumatoid arthritis, ankylosing spondylitis, asthma and so on. The study on β-arrestins will reveal the mechanisms of inflammatory diseases and provide new strategy for clinical treatment. Here we reviewed the recent progress on the role of β-arrestins in regulating the inflammatory diseases.
