大鼠急性颈脊髓损伤后体内P物质含量变化与黄芪多糖的关系 被引量:1

摘要 [目的]探讨黄芪多糖对大鼠急性颈脊髓损伤后血清及肺泡灌洗液中P物质含量变化的影响。[方法]成年wistar大鼠80只,体重240~250g,雌雄不限。大鼠随机分为四组,分别为黄芪多糖组、等渗盐水组、空白对照组、假手术组,每组20只。各组按取材时间不同又分为造模后24h、3d、1周、2周共4个时间点,每个时间点5只大鼠。采用C7段脊髓改良Allen’s打击法制作大鼠脊髓损伤模型,打击力度为10×2.5g·cm;假手术组仅暴露C7段脊髓。造模后等渗盐水组大鼠肌肉注射生理盐水0.5ml,1次/d,黄芪多糖组大鼠肌肉注射黄芪多糖注射液0.5ml,1次/d;其余2组未注射任何液体。[结果]在不同时间点处死大鼠,测量各时间点每组大鼠血清和肺泡灌洗液P物质含量。结果显示黄芪多糖对脊髓损伤大鼠P物质含量变化有影响。伤后各时间点黄芪多糖组血清及肺泡灌洗液P物质含量较空白对照组及等渗盐水组为低。黄芪多糖组脊髓损伤后3d、1周、2周血清及伤后3d肺泡灌洗液P物质含量较空白对照组和等渗盐水组有统计学差异(P〈0.05)。[结论]黄芪多糖可以减少大鼠急性颈脊髓损伤后血清及肺泡灌洗液中P物质的含量。这一结果可能对治疗急性脊髓损伤后肺水肿有帮助。 [ Objective] To investigate the effect of astragalus in acute cervical spinal cord injured (SCI) rats. [ Method ] Eighty Wistar rats (240 -250 g) were treated with astragalus polysaccharide (APS), and substance P (SP) levels were measured at different time points. The rats were randomly divided into 4 groups ( APS, normal saline, blank, and sham groups) , which were subdivided into groups based on time after SCI (24 hours,3 days,1 week,and 2 weeks). SCI was induced at the level of seventh cervical vertebrae ( C7 ) by dropping a weight of 10 g from a height of 25 mm ( Allen' s model). In the sham group, only the laminas were removed,leaving the spinal cord intact at C7. The respective homologous liquids were slowly injected semel in die into the gluteus of SCI rats in the APS and normal saline groups. [ Result] All of the animals were sacrificed at different time points ( 24 hours ,3 days, 1 week,2 weeks) after injury. The levels of SP in the serum and bronchoalveolar lavage fluid (BALF) were measured. The levels of SP in the serum and BALF were lower in the APS group than in the normal saline and blank groups at each time point, particularly at 3 days, and 1 and 2 weeks in the serum and at 3 days in the BALF after SCI ( P 〈 0. 05 ). [ Conclusion ]APS can reduce the levels of SP in the serum and BALF in acute cervical SCI rats. Therefore,it may be a useful treatment for pulmonary edema in SCI patients.
