组织浸润的T-bet^+淋巴细胞与胃癌发生发展关系研究

Preliminary investigation into the distribution of subsets of tumor infiltrating T-bet^+ lymphocytes in human gastric cancer tissues

摘要 研究胃癌组织中浸润T-bet+淋巴细胞的亚群分布。采用免疫荧光标记及流式细胞术,检测胃癌患者外周血单个核细胞、胃癌及癌旁正常组织中浸润淋巴细胞中T-bet+淋巴细胞的亚群分布。流式检测结果表明,外周血单个核细胞中,Tbet在CD4+T细胞、CD8+T细胞及CD56+NK细胞中均有表达,经过分离浸润淋巴细胞及荧光抗体标记后,流式检测结果表明胃癌及癌旁正常组织中,T-bet在浸润的CD4+T细胞、CD8+T细胞及CD56+NK细胞中亦有表达,并且在CD8+T细胞及CD56+NK细胞中表达水平较高。结果表明,胃癌组织中浸润T-bet+淋巴细胞包括CD4+T、CD8+T细胞及CD56+NK细胞等,提示在胃癌发生发展中T-bet参与调控抗肿瘤细胞免疫应答。 To investigate the subset distribution of infiltrating T-bet+ lymphocytes in human gastric cancer tissues, immunoflu- orescence staining and flow eytometry were used to detect the subset distribution of T-bet+ lymphocytes in human peripheral blood mononuclear cells, infiltrating lymphocytes in gastric cancer tissues as well as in the adjacent normal tissues. Flow cy- tometry showed that T-bet expression could be found in CD4+ T cells, CD8+ T cells and CD56+ NK cells both in the PMNC and in the infiltrating lymphocytes. In the infiltrating lymphocytes, high expression of T-bet could be found in CD8+ T cells and CD56+ NK cells. These findings indicate that the infiltrating T-bet+ lymphocytes in human gastric cancer tissues are composed of CD4+ T cells, CD8+ T cells and CD56+ NK cells, suggesting that T-bet might be involved in the regulation of cell-mediated anti-tumor response in the progression of human gastric cancer.
