摘要 为了筛选最佳的杜仲叶片光合色素含量测定方法,以浸提和研磨2种方法,分别用4种浸提液进行提取,并用紫外分光光度计对各组合不同时间光合色素含量进行测定.结果表明,研磨法显著优于浸提法,浸提速度最快的是丙酮∶乙醇∶水=4.5∶4.5∶1浸提液,其次为丙酮∶乙醇=1∶1浸提液,但丙酮+乙醇+水浸提液易与叶片中某种物质发生反应而使浸提物变黑.研磨法各浸提液所得光合色素浓度在12 h内无显著差异.杜仲叶片光合色素含量最佳提取方法为研磨法;浸提液为丙酮∶乙醇=1∶1,测定时间在12 h内. In order to select the optimal assay method for testing photosynthetic pigment content in leaves of Eucommia ulmoides, extraction and grinding method were applied to extract the pigment by using four kinds of solvent. The content of photosynthetic pigment was then determined by ultraviolet spectrophotometer. Our results showed that the grinding method was significantly effective than extraction method. The solvent of acetone: ethanol: water in ratio of 4.5 : 4.5 : 1 was fastest for extractions, the next was acetone-ethanol (1:1 ). However, the solvent of acetone, ethanol and water could react with components in leaves and the extracts turned into black. There were no significant differences among concentrations of photosynthetic pigment extracted by grinding method with different kinds of solvent within 12 hours. In conclusion, grinding method was optimal for extracting photosynthetic pigment from Eucommia leaves. , the solvent was acetone-ethanol 1:1 ) and the extracts should be tested within 12 hours.
