摘要
目的:研究用AB-8型大孔树脂富集纯化川续断中川续断皂苷Ⅵ的最佳工艺。方法:以川续断皂苷Ⅵ的动、静态吸附量和解吸附率为指标,考察了AB-8型大孔树脂对川续断中川续断皂苷Ⅵ的吸附和解吸附性能。结果:AB-8型大孔树脂纯化川续断皂苷Ⅵ的最佳工艺为:上样溶液的浓度为固含量0.08g/ml,1g树脂上样量相当于1.2g生药材,先用30%乙醇除杂,再用70%乙醇解吸附,用量皆为3个柱体积,流速200ml/h。得到富集浸膏的得率为8.93%,浸膏中川续断皂苷Ⅵ的含量为(65.32±1.73)%,转移率为95%。结论:该工艺能有效纯化、富集川续断药材中的川续断皂苷Ⅵ。
Objective:To study the optimal purification process for gathering and purifying asperosaponin Ⅵ from Dipsacus asper Wall.with AB-8 macroporous resin.Methods:With the dynamic and static absorption and desorption rate of asperosaponin Ⅵ as indices,the performance of asperosaponin Ⅵ from Dipsacus asper Wall.was investigated with AB-8 macroporous resin.Results:The optimal purification process of asperosaponin Ⅵ from Dipsacus asper Wall.was as follows:solid content of sample solution was 0.08 g/ml,absorption capacity of 1 g macroporous resin was 1.2 g crude drug.Then,30% and 70% ethanol solutions were used to remove impurities and desorb asperosaponin Ⅵ,all with a volume of 3 columns and a flow rate of 200 ml/h.The yield of enriched extract was 8.93%,the content of asperosaponin Ⅵ in the extract was (65.32±1.73) %,and diversion rate of asperosaponin Ⅵ was 95 %,Conclusion:The purification process could effectively purify asperosaponin Ⅵ from Dipsacus asper Wall..
