白刺果实水溶性多糖的单糖组成研究 被引量:5

摘要 以白刺果实为原料,采用热水浸提法提取水溶性多糖,并对水溶性多糖的单糖组成进行研究。石油醚脱脂后,采用80℃热水浸提法提取果粉中的水溶性糖,料液比1︰10(g/mL),提取2 h,重复提取3次,合并提取液后减压浓缩至原体积的1/3,加入3倍体积95%的乙醇,4℃条件下静置24 h,沉淀即为水溶性多糖,采用Sevage法除蛋白,反复醇洗脱色,经低温干燥得到水溶性多糖。经过柱前衍生HPLC法分析白刺果实水溶性多糖的单糖组成。结果表明:水溶性多糖得率约为1%。白刺果实水溶性多糖的单糖由葡萄糖、鼠李糖、阿拉伯糖、半乳糖、半乳糖醛酸、甘露糖6种单糖组成,单糖组成质量比为甘露糖︰鼠李糖︰半乳糖醛酸︰葡萄糖︰半乳糖︰阿拉伯糖=9.2︰3.3︰1.1︰1︰1.9︰2.3。单糖组成中葡萄糖所占比例最低,半乳糖醛酸与葡萄糖含量相近,甘露糖所占比例最高,是葡萄糖的9倍多,半乳糖所占比例约为葡萄糖的2倍,阿拉伯糖所占比例是葡萄糖的2.3倍。 Useing the Nitraria fruit as raw material, the water-soluble polysaccharides was exlracted by hot water, and composition of polysaccharide were studied. Degreasing with petroleum ether, by 80℃ hot water extraction of the water-soluble sugar powder, liqtfid ratio 1 : 10 (g/mL), extraction 2 h, repeated extracting for 3 times, merging the extracted 1/3 is decompression concentrated to the original volume of liquid, adding 3 times volume of 95% ethanol, static 24 h under the condition of 4℃, precipitation is water- soluble polysaccharide, removing protein by Sevage method, repeated alcohol washed bleaching, low-temp drying to obtain water soluble polysaccharide. Analyze the monosaccharide composition of nitraria fruit water soluble polysaccharide by precolumn derivatization HPLC. The results showed that yield of water-soluble polysaccharide are about 1%. Monosaccharide nitraria fiuit water soluble polysaccharide is composed of glucose, rhamnose, glucose, galactose, Arabia galacturonic acid, mannose 6 monosaccharide composition, monosaccharide composition ratio of mannose : rhamnose : galacturonic acid glucose : galactose : Arabia sugar=9.2 : 3.3 : 1.1 : 1 : 1.9 : 2.3. Monosaccharide composition of glucose in the lowest proportion, galacturonic acid and glucose were similar, mannose accounted for the highest pmpoaion, more than nine times of glucose, galactose accounted for about two times of glucose, Arabia sugar accounts for the proportion is 2.3 times of glucose.
