摘要
为了研究外源性孕酮对牛卵母细胞体外核成熟的作用,通过向成熟培养液中添加10μg/mL FSH+10μg/mL LH(对照组)和0、10、100、1000 ng/mL的外源性孕酮,对牛卵丘卵母细胞复合体进行体外成熟培养,在培养8 h后用醋酸地衣红染色观察各自的生发泡破裂率,然后在成熟22 h统计第一极体率,体外受精后7~8天统计囊胚率。结果表明:对照组卵母细胞的生发泡破裂率(75.0%)和极体率(79.6%)均明显高于添加不同浓度孕酮的各处理组(P〈0.05),而囊胚率各组之间均无显著差异(P〉0.05)。因此,外源性孕酮不能促进牛卵母细胞的核成熟,单独添加外源性孕酮不能起到与内源性孕酮相同的作用。
In this experiment,it was aimed to investigate the effect of exogenous progesterone on in vitro nuclear maturation of bovine oocytes through adding with 10 μg/mL FSH + 10 μg/mL LH(control),0,10,100,1000 ng/mL exogenous progesterone respectively into the in vitro maturation medium for bovine cumulus-oocyte complexes(COCs).The germinal vesicle breakdown(GVBD) rate of each group after 8 h using acetic acid-orcein staining,and counted first polarbody(PbI) rate after 22 h were all analyzed.Finally,the blastocyst rate after 7-8 days of in vitro fertilization was also counted.The results showed that,GVBD(75.0%) and PbI(79.6%) rate of the control group were significantly higher than the other ones adding with exogenous progesterone(P<0.05),and blastocyst rate of each group didn’ t show significant variation(P>0.05).Therefore,it could be concluded that the exogenous progesterone could not play a role for improving the nuclear maturation of bovine oocytes,it also meant that exogenous progesterone alone could not take the same effect as endogenous progesterone in such kind of processes.
