摘要
目的观察祛风宣肺方对豚鼠咳嗽模型咳嗽次数及气道反应性的影响。方法选用SPF级Hartley雄性豚鼠40只,随机分为正常对照组、模型组、中药组、西药组,除正常对照组外,其余各组动物腹腔内注入免疫抑制剂环磷酰胺(30 mg/kg),2 d后腹腔注入卵蛋白2 mg和氢氧化铝100mg,3周后腹腔注射0.01 mg卵蛋白和100 mg氢氧化铝以加强免疫反应,使豚鼠致敏。又3周后将其置于自制雾化箱内给予10 g/L卵蛋白溶液雾化吸入60 s激发咳嗽,造模成功。中药组给予祛风宣肺方口服,西药组口服孟鲁司特钠片,连续给药10 d后,测定各组咳嗽次数、气道反应性、支气管肺泡灌洗液白细胞总数及分类。结果咳嗽次数方面,模型组与正常对照组相比,咳嗽次数明显增多(P<0.05),中药组及西药组与模型组相比咳嗽次数显著减少(P<0.05),中药组与西药组相比咳嗽次数减少(P<0.05)。气道反应性方面,激发后各组气道阻力均较前上升,模型组显著高于其他组(P<0.05),中药组及西药组较模型组显著偏低(P<0.05),中药组明显低于西药组(P<0.05)。支气管肺泡灌洗液白细胞总数及分类方面,模型组与正常对照组相比,白细胞计数、单核细胞及嗜酸性粒细胞显著增多,中药组及西药组与模型组相比以上细胞数减少,中药组与西药组相比以上细胞数减少(P<0.05)。结论祛风宣肺方对豚鼠咳嗽模型有较好的治疗作用,作用机制可能与减轻气道炎症、进而降低气道高反应有关。
Objective To observe the influences of Qufeng Xuanfei Fang on cough and airway responsiveness in guinea pigs with airway hypersensitivity.Methods Male Hartley guinea pigs (n =40) were intraperitoneally injected cyclophosphamide (30 mg/kg),and ovalbumin (2 mg) and aluminium hydroxide (100 mg/kg) 2 days later,and ovalbumin (0.01 mg) and aluminium hydroxide (100 mg/ kg) 3 weeks later for boosting immunization and sensitization.After 3 weeks,guinea pigs were given aerosol inhalation of ovalbumin solution (10 mg/mL) for 60 seconds to provoke cough.All guinea pigs were divided into control group,model group,Chinese medicinal group and Western medicinal group.Chinese medicinal group was orally given Qufeng Xuanfei Fang and Western medicinal group was orally given montelukast sodium for continuous 10 days.Then counting of cough,airway responsiveness,and counting and differentiation of white blood cell (WBC) in bronchoalveolar lavage fluid were detected in all groups.Results Counting of cough increased significantly in model group compared with control group (P < 0.05),decreased in Chinese medicine group and montelukast sodium group compared with model group (P < 0.05),and decreased in Chinese medicinal group compared with Western medicinal group (P < 0.05).Airway resistance in all groups increased after provoking cough.The value of airway resistance in model group was relatively higher than those in other groups (P < 0.05),was lower in Chinese medicinal group and Western medicinal group than that in other groups (P < 0.05),and was lower in Chinese medicinal group than that in Western medicinal group (P < 0.05).In the aspect of counting and differentiation of WBC in bronchoalveolar lavage fluid,total cells counting,monocyte counting and eosinophils counting were more in model group than those in control group (P < 0.05),were less in Chinese medicinal group and Western medicinal group than those in model group (P < 0.05),and were less in Chinese medicinal group than those in Western medicinal group (P < 0.05).Conclusion Qufeng Xuanfei Fang has a satisfactory curative effect on guinea pigs with airway hypersensitivity,and the mechanism may be related to alleviating airway inflammation and relieving airway hypersensitivity.
